Article Abstract

As the rate-limiting enzyme in Glutathione (GSH) biosynthesis, the mammalian glutamate cysteine ligase (Gcl) catalytic (Gclc) and modifier (Gclm) subunits are regulated at multiple levels, whereas the function and regulatory mechanism of insect Gcl remain to be explored. In this study, we identified and characterized and in . and were highly expressed in the hindgut and relatively less expressed in other tissues. The exposure of the third instar larvae to LC of emamectin benzoate (EMB) significantly reduced the GSH content with a concomitant upregulation of and Further pretreatment with L-BSO, the Gcl inhibitor, increased the susceptibility of to EMB. Consistently, overexpression of and increased the Sf9 cell viability under EMB treatment. Finally, both RNAi and the dual-luciferase reporter assay in Sf9 cells revealed that is regulated by transcription factor CncC. These data provide insights into the function and regulatory mechanism of insect Gcl, and they imply that disruption of the redox homeostasis might be a practical strategy to enhance the insecticidal activity of EMB and other insecticides.

Flavin-based electron bifurcation (FBEB) is employed by microorganisms for controlling pools of redox equivalents by reversibly splitting electron pairs into high- and low-energy levels from an initial midpoint potential. Our ability to harness this phenomenon is crucial for biocatalytic design which is limited by our understanding of energy coupling in the bifurcation system. In Pyrococcus furiosus, FBEB is carried out by the NADH-dependent ferredoxin:NADP-oxidoreductase (NfnSL), coupling the uphill reduction of ferredoxin in NfnL to the downhill reduction of NAD in NfnS from oxidation of NADPH.

The present study aimed to determine the effects of potato cultivars, processing methods, and processing conditions on acrylamide formation in fresh-cut potato chips. Tubers of five potato cultivars (Daxiyang, Shiyan1, V7, Youjin885, Zao35) were processed into potato chips using five methods (pan-cooking, deep-frying, oven-baking, microwaving, and air-frying). Among the five cultivars, tubers of potato Youjin885 were identified as the most suitable cultivar for producing potato chips with the lowest acrylamide and its precursor contents.

The systemic administration of low-molecular-weight heparin (LMWH) as the standard initial treatment for venous thromboembolism is intended to prevent thrombus growth during the acute phase and prevent thrombus recurrence during the chronic phase. However, precisely controlling the specificity and localization of LMWH presents significant challenges, which has necessitated research into targeted delivery methods. Furthermore, due to different thrombosis mechanisms, tailored pharmacologic interventions are needed, particularly for arterial and venous thrombosis.
