Background: Studies examining the association of chronic kidney disease (CKD) with cancer risk have demonstrated conflicting results.
Methods: This was an individual participant data meta-analysis including 54 international cohorts contributing to the CKD Prognosis Consortium. Included cohorts had data on albuminuria [urine albumin-to-creatinine ratio (ACR)], estimated glomerular filtration rate (eGFR), overall and site-specific cancer incidence, and established risk factors for cancer.

Cardiovascular-kidney-metabolic (CKM) syndrome is a term that is increasingly used to describe interconnected conditions that lead to poor health outcomes, including cardiovascular disease, chronic kidney disease, type 2 diabetes, and obesity. Historically, there have been very few targeted pharmacotherapies available that have changed cardiovascular outcomes for people with CKM syndromes; however, over the past decade, new pharmacologic options have rapidly expanded, with strong evidence for cardiovascular and kidney protective benefits in CKM conditions. Of note, sodium-glucose cotransporter-2 inhibitors and glucagon-like peptide-1 receptor agonists have emerged as key therapeutic options and are now widely guideline-endorsed.

Chronic kidney disease of unknown etiology has been reported in Mesoamerican regions and other parts of the world, with increasing evidence pointing to heat stress as a central contributing factor. The incidence of acute kidney injury appears to correlate strongly with heat exposure, as demonstrated in both human and animal studies. The underlying mechanisms of heat-induced kidney injury are likely multifactorial, involving hemodynamic changes, immune responses, and possibly coagulopathies.

Circulating levels of 3-carboxy-4-methyl-5-propyl-2-furanpropionic acid (CMPF), a metabolite derived from dietary furan fatty acids primarily found in marine food sources, have long been recognized as biomarkers for fish intake. However, elevated CMPF levels are also observed in patients with type 2 diabetes or chronic kidney disease and in healthy people associated with a reduced infection risk, suggesting potential bioactive roles in metabolism and immune function. Yet, the possible causal mechanisms behind these associations are unknown.

Metabolic dysfunction-associated steatohepatitis (MASH) affects a large proportion of the global population and is widely regarded as the fastest growing cause of hepatocellular carcinoma. Currently, approved therapeutic strategies for MASH are limited. Therefore, this study used the Connectivity Map (CMap) database to identify a candidate compound for MASH, evaluate its efficacy in experimental models, and explore its mechanism of action.
