Three new rearranged diterpenoids, strophioblachins A-C (-), eight new diterpenoids, strophioblachins D-K (-), and seven previously described diterpenoids (-) were purified from the aerial parts of Compounds and contain a rare 6/6/5/6 ring system, while has an uncommon tricyclo[4.4.0.0]tridecane-bridged unit, and their diterpenoid skeletons are being reported for the first time. Utilizing spectroscopic and HRESIMS data analysis, the structures of the new compounds (-) were established, and ECD and C NMR calculations were used to confirm the relative and absolute configurations of and . The absolute configurations of compounds , , and were established using single-crystal X-ray diffraction. The results of testing for anticardiac hypertrophic activity demonstrated that compounds and dose-dependently lowered the mRNA expression of and . Protein levels were confirmed by Western blotting, which also demonstrated that compounds and lowered the expression of the hypertrophic marker ANP. The cytotoxic activity against neonatal rat cardiomyocytes was assayed in vitro by the CCK-8 and ELISA methods, and the results showed that compounds and were only very weakly active in the range.

Background: Hypertrophic Cardiomyopathy (HCM) is an inherited heart disease and the pathogenesis of HCM involves genetic mutations, hemodynamic stress, and metabolic factors, with myocardial fibrosis playing a crucial role in severe clinical events. IL-33/ST2 signaling pathway known for its roles in immune response and tissue repair, participates in cardiac protection and anti-cardiac fibrosis in heart failure. The role of in HCM remains unclear, and IL-33/ST2 pathway and broader inflammatory responses may be critical in HCM.

Seven new β-triketone-acylphloroglucinol-monoterpenoid hybrids, namely callistevimones A-G (1-7), were isolated from Callistemon viminalis fruits. Their structures and absolute stereochemistry were accomplished through a comprehensive analytical method involving mass spectrometry, NMR, ECD calculation, QM-NMR calculation, and single-crystal X-ray crystallography. Compounds 1 and 2 are first examples of β-triketone-acylphloroglucinol-phellandrene with an enlarged-ring.

In the present investigation, fourteen unprecedented podocarpane diterpenoids strophiolosas A-J, L-N and P (1-10, 12-14, 16), two new cleistanthane derivatives strophiolosas Q-R (17-18), two new dibenzopyroan-ones and one new tetralone strophiolosas S-U (19-21), were isolated from the whole plant of Strophioblachia glandulosa.
