Lipotoxicity induced by the overload of lipid in the liver, especially excess free cholesterol (FC), has been recognized as one of driving factors in the transition from non-alcoholic fatty liver (NAFL) to non-alcoholic steatohepatitis (NASH). MicroRNA (miR)-379-5p has been reported to play regulatory roles in hepatic triglyceride homeostasis, but the relationship of miR-379-5p and hepatic cholesterol homeostasis has never been touched. In the current study, we found that hepatic miR-379-5p levels were decreased obviously in NAFLD patients and model mice compared with their controls. Moreover, miR-379-5p was discovered to be able to inhibit intracellular FC accumulation and alleviate mitochondrial damage induced by palmitic acid (PA) in vitro. Furthermore, overexpression of miR-379-5p in HFHC-fed db/db mice could reduce the level of hepatic total cholesterol (TC) and FC, and ameliorate hepatic injury reflected by the lower serum alanine aminotransferase (ALT) and aspartate transaminase (AST). Subsequently, by combining spectrometry (MS) and luciferase assay, we identified miR-379-5p suppressed STAT1 through transcriptional and translational regulation. Finally, we confirmed that STAT1 was a transcriptional factor of HMGCS1. In conclusion, miR-379-5p inhibits STAT1 expression and regulates cholesterol metabolism through the STAT1/HMGCS1 axis, suggesting miR-379-5p might be applied to improve lipotoxicity in the future.

Brillouin microscopy allows mechanical investigations of biological materials at the subcellular level and can be integrated with Raman spectroscopy for simultaneous chemical mapping, thus enabling a more comprehensive interpretation of biomechanics. The present study investigates different in vitro glioblastoma models using a combination of Brillouin and Raman microspectroscopy. Spheroids of the U87-MG cell line and two patient-derived cell lines as well as patient-derived organoids were used.

Deciphering the multicomponent of cell membranes at the single-cell level is critical for understanding pathological mechanisms such as tumor metastasis, yet remains technically daunting due to the membrane's nanoscale thickness and ultralow molecular abundance. Here, we introduce a surface-assisted vacuum ultraviolet laser desorption-ionization mass spectrometry imaging (SAVUVDI-MSI) platform that overcomes long-standing challenges of cytoplasmic interference and insufficient sensitivity. Leveraging the nanoscale depth profiling capability of VUV-LDI, we achieve precise ablation of a single-cell membrane.

Previously, changes in MG lipidomes induced by inactivation of critical genes of meibogenesis, such as and others, were shown to cause MG-dysfunction- and dry eye-like signs in mice. Here, we describe the impact of the lipid composition of meibum on its physiological properties, specifically thermotropic/melting characteristics, using various wild type and mutant animals, and compare them with meibum of healthy humans and patients with abnormal meibum.

Objective: To investigate the association between thyroid hormone sensitivity indices and bone metabolism markers in newly diagnosed middle-aged and elderly type 2 diabetes mellitus (T2DM) patients with normal thyroid function.
Method: We retrospectively analyzed 350 newly diagnosed T2DM patients (≥ 45 years), stratified by bone mineral density into Group A (normal bone density group) and Group B (low bone mass and osteoporosis group). General data and clinical biochemical parameters were collected: free triiodothyronine (FT3), free thyroxine (FT4), thyroid-stimulating hormone (TSH), 25-hydroxyvitamin D (25(OH)D), parathyroid hormone (PTH), osteocalcin (OC), bone-specific alkaline phosphatase (BALP), serum calcium (Ca), serum phosphorus (P), fasting plasma glucose (FPG), glycosylated hemoglobinA1c (HbA1c), total cholesterol (TC), triglyceride (TG), low-density lipoprotein cholesterol (LDL-C), high-density lipoprotein cholesterol (HDL-C), serum creatinine (SCr), serum uric acid (SUA), and estimated Glomerular Filtration Rate (eGFR).
