Article Abstract

Probing endogenous molecular profiles in living entities is of fundamental significance to decipher biological functions and exploit novel theranostics. Despite programmable nucleic acid-based aptasensing systems across the breadth of molecular imaging, an aptasensing system enabling in vivo imaging with high sensitivity, accuracy, and adaptability is highly required yet is still in its infancy. Artificial catalytic DNA circuits that can modularly integrate to generate multiple outputs from a single input in an isothermal autonomous manner, have supplemented powerful toolkits for intracellular biosensing research. Herein, a multilayer nonenzymatic catalytic DNA circuits-based aptasensing system is devised for in situ imaging of a bioactive molecule in living mice by assembling branched DNA copolymers with high-molecular-weight and high-signal-gain based on avalanche-mimicking hybridization chain reactions (HCRs). The HCRs aptasensing circuit performs as a general and powerful sensing platform for precise analysis of a series of bioactive molecules due to its inherent rich recognition repertoire and hierarchical reaction accelerations. With tumor-targeting capsule encapsulation, the HCRs aptasensing circuit is specifically delivered into tumor cells and allowed the high-contrast imaging of intracellular adenosine triphosphate in living mice, highlighting its potential for visualizing these clinically important biomolecules and for studying the associated physiological processes.

It is imperative to develop convenient methodology to enable the simultaneous detection of enrofloxacin (ENR) residue markers coexisted in the aquatic environment. To this end, an efficient dual-channel photoelectrochemical (PEC) aptasensing approach is presented for the detection of trace ENR residue markers simultaneously through a single signal acquisition. Primarily, cathodic PEC aptasensing platform with superior anti-interference capability and high sensitivity was designed and constructed on the basis of the high visible light-harvesting CuInS photocathode and BiS/CN photoanode; and the simultaneous detection of ENR residue markers was then realized by the spatial resolution of aptasensing interfaces.

Accurate diagnosis of Gram-negative bacterial infections hinges critically on endotoxin detection, with ultrasensitive quantification playing a pivotal role in clinical diagnosis, precision therapy, food safety and environmental monitoring. Herein, we pioneer an electrochemical aptasensing platform based on metal-free photoinduced ring-opening metathesis polymerization (MF photo-ROMP) for endotoxin analysis. Specifically, the aptamer is employed for selective capture of endotoxin, whose glycan chain is then modified with 4-formylphenylboronic acid via boronate affinity.

The escalating crisis of hospital-acquired multidrug-resistant (MDR) infections, particularly carbapenemase 2-expressing (KPC-2 KP) and MDR- (AB), demands rapid diagnostic solutions. We developed a dual nanozyme-powered colorimetric aptasensor leveraging a cascade amplification mechanism, a metal-organic framework (MOF)-on-MOF nanostructure with peroxidase-like activity. Cu-MOF@PMOF(Fe) integrates catechol oxidase-like activity, with the Cu-MOF core oxidizing catechol to generate HO, and the PMOF(Fe) shell utilizes HO to oxidize the TMB substrate, producing dual-wavelength signals at 370 and 652 nm for ultrasensitive detection.

Herein, an integrated ultraswelling microneedle aptamer-recognition tester (uSMART) was developed for POC monitoring of antibiotic-resistant bacterial infections and therapeutic outcomes. The hyaluronic acid methacryloyl/sodium hyaluronate-microneedle (HAMA/SH-MN) module with excellent swelling and mechanical properties could cross the epidermis and access enough dermis ISF in a minimally invasive, painless, and rapid manner.

Regulating the microscale carrier transport in semiconductor materials to enhance the macroscopic photoelectrochemical properties would drive technological advancement and industrial upgrading. In this work, we integrated the maturity of microfluidic technology control methods with current photoelectrochemical analysis sensing techniques to establish a microfluidic photoelectrochemical induction system by modulating the photon-to-current efficiency under zero bias. Traditionally, target recognition has been achieved by affecting the transport of carriers through the space resistance equivalent of the solid-liquid contact interface, which is limited in sensitivity enhancement.
