Ruthenium red (RR) inhibits calcium (Ca) entry from the cytoplasm to the mitochondria, and is involved in maintenance of Ca homeostasis in mammalian cells. Ca homeostasis is very important for further embryonic development of fertilized oocytes. However, the effect of RR on mitochondria-Ca (mito-Ca) levels during in vitro fertilization (IVF) on subsequent blastocyst developmental capacity in porcine is unclear. The present study explored the regulation of mito-Ca levels using RR and/or histamine in fertilized oocytes and their influence on blastocyst developmental capacity in pigs. Red fluorescence intensity by the mito-Ca detection dye Rhod-2 was significantly increased (P < 0.05) in zygotes 6 h after IVF compared to mature oocytes. Based on these results, we investigated the changes in mito-Ca by RR (10 and 20 μM) in presumptive zygotes using Rhod-2 staining and mito-Ca uptake 1 (MICU1) protein levels as an indicator of mito-Ca uptake using western blot analysis. As expected, RR-treated zygotes displayed decreased protein levels of MICU1 and Rhod-2 red fluorescence intensity compared to non-treated zygotes 6 h after IVF. Blastocyst development rate of 20 μM RR-treated zygotes was significantly increased 6 h after IVF (P < 0.05) due to improved mitochondrial functions. Conversely, the blastocyst development rate was significantly decreased in histamine (mito-Ca inhibitor, 100 nM) treated zygotes (P < 0.05). The collective results demonstrate that RR improves blastocyst development in porcine embryos by regulating mito-Ca and MICU1 expression following IVF.

Introduction: embryo production (IVP) in the domestic cat () remains highly variable owing to intrinsic reproductive traits and the absence of fully standardized protocols.
Methods: We retrospectively analysed 108 IVP replicates produced under a single protocol (2020-2024) to quantify the effects of season, donor age and methodological parameters on oocyte yield and embryo development.
Results: Winter proved the most favourable season for both oocyte recovery and blastocyst formation, whereas spring, despite lower initial yields, achieved the greatest post-selection oocyte retention.

RNA-binding proteins (RBPs) are critical regulators of post-transcriptional gene expression and RNA processing during mammalian oocyte development. SERPINE1 mRNA-binding protein 1 (SERBP1), a conserved RNA-binding protein (RBP), exhibits prominent expression in the female reproductive system and throughout oogenesis. Conditional deletion of Serbp1 using oocyte-specific Zp3/Gdf9-Cre drivers resulted in arrested oocyte growth, female infertility, and failure of blastocyst formation from two-cell embryos.

Purpose: In piezo-ICSI, the first polar body (PB) of a metaphase II (MII) oocyte is generally oriented in the 6 or 12 o'clock position relative to sperm injection at 3 o'clock. However, the ooplasmic cell membrane may be damaged during drilling of the zona pellucida by piezo pulses. Here, we tested a new piezo-ICSI method in which the PB is set at the 2 or 4 o'clock position, so that zona drilling is performed through the widest position in the perivitelline space (para-PB piezo).

Serotonin (5-hydroxytryptamine; 5-HT) is transported into the human placenta through the serotonin transporter (SERT/SLC6A4) on the surface of the syncytiotrophoblast. During this transit, a significant amount of 5-HT becomes concentrated in the cytotrophoblast nucleus. We used immunochemistry, inhibitors of SERT and transglutaminase 2, and RNA sequencing to elucidate the mechanism and consequences of this nuclear localization.
