Article Abstract

In plants, transcripts move to distant body parts to potentially act as systemic signals regulating development and growth. Thousands of messenger RNAs (mRNAs) are transported across graft junctions via the phloem to distinct plant parts. Little is known regarding features, structural motifs, and potential base modifications of transported transcripts and how these may affect their mobility. We identified Arabidopsis thaliana mRNAs harboring the modified base 5-methylcytosine (mC) and found that these are significantly enriched in mRNAs previously described as mobile, moving over graft junctions to distinct plant parts. We confirm this finding with graft-mobile methylated mRNAs TRANSLATIONALLY CONTROLLED TUMOR PROTEIN 1 (TCTP1) and HEAT SHOCK COGNATE PROTEIN 70.1 (HSC70.1), whose mRNA transport is diminished in mutants deficient in mC mRNA methylation. Together, our results point toward an essential role of cytosine methylation in systemic mRNA mobility in plants and that TCTP1 mRNA mobility is required for its signaling function.

Epithelioid hemangioendothelioma (EHE) is an exceptionally rare vascular tumor with an incidence of <1/million people/year. This case report describes a patient who initially presented with progressive left-sided neck swelling and dysphagia and was found to have EHE at the confluence of the left subclavian and internal jugular veins. Diagnosis was confirmed with a core needle biopsy after an initial inconclusive fine-needle aspiration.

Peripheral nerve defect repair is a complex process that involves multiple cell types; perineurial cells play a pivotal role. Hair follicle neural crest stem cells promote perineurial cell proliferation and migration via paracrine signaling; however, their clinical applications are limited by potential risks such as tumorigenesis and xenogeneic immune rejection, which are similar to the risks associated with other stem cell transplantations. The present study therefore focuses on small extracellular vesicles derived from hair follicle neural crest stem cells, which preserve the bioactive properties of the parent cells while avoiding the transplantation-associated risks.

Background: Neoadjuvant therapy can reduce the size of gastroesophageal tumors to the extent that they are no longer macroscopically visible. This may increase the risk of microscopic-positive resection margins. One potential method to reduce this uncertainty could be the preoperative endoscopic marking of proximal tumor margins with BioXmark, a novel liquid fiducial marker.
