Article Abstract

The quantitative real-time polymerase chain reaction (qPCR) is one of the most commonly molecular methods used today. It is central to numerous assays that have since been developed and described around its optimization. The qPCR assay has been studied in great detail and due to its comprehensive knowledge, excellent performance (sensitivity of one single copy), and internal amplification control, it represents a suitable test platform for qPCR examinations. In this study, we compared ten different polymerases (or ready-to-use mastermixes) as possible (economic) alternatives to our gold standard Platinum polymerase. We sought to determine the reproducibility of these assays under modified conditions, which are realistic because published assays are frequently used with substituted polymerases. Surprisingly, there was no amplification at all with some of the tested polymerases, even although the internal amplification control worked well. Since adaptation of the thermal profile and of MgCl concentration could restore amplification, simple replacement of the polymerase can destroy a well-established assay leading up to >10-fold less analytical sensitivity. Further, validation using Poisson and PCR-Stop analyses revealed limits to some assay-polymerase combinations and emphasize the importance of validation.

Extreme event attribution assesses how climate change affected climate extremes, but typically focuses on single events. Furthermore, these attributions rarely quantify the extent to which anthropogenic actors have contributed to these events. Here we show that climate change made 213 historical heatwaves reported over 2000-2023 more likely and more intense, to which each of the 180 carbon majors (fossil fuel and cement producers) substantially contributed.

The potential of PM to cause lung cancer has been well established; however, evidence regarding which specific components are responsible remains limited. We investigated dissolved organic matter (DOM) in PM using high-resolution mass spectrometry (HRMS) and cellular DNA damage assays to elucidate molecular composition and sources of carcinogenic components. Our analysis revealed hundreds of genotoxic compounds, with condensed aromatic amines predominating in number, abundance, and contribution to overall genotoxicity.

The controlled atmosphere (CA) technique, though well-established in perishable food preservation, had been underexplored in broth production. This study investigated the effects of varying cooking times (30, 60, 90 min) and temperatures (105 °C, 110 °C, 115 °C) under CA and non-controlled atmosphere (NCA) conditions on the sensory, compositional, flavor, and stability characteristics of pork bone broth. Clear differences between CA and NCA were found and analyzed.

Topic: The aim of this study was to assess the meta-analysis of the studies comparing transepithelial photorefractive keratectomy (TransPRK) to classical photorefractive keratectomy (PRK) (mechanical or alcohol-assisted).
