A rapid and specific liquid chromatography-tandem mass spectrometry method with a wide linear range was developed and validated for the simultaneous quantification of Vitamin K1 (VK1) trans- and cis- isomers in human plasma. Background: Tumor heterogeneity and antigen escape are mechanisms of resistance to chimeric antigen receptor (CAR)-T cell therapy, especially in solid tumors. Targeting multiple antigens with a unique CAR construct could be a strategy for a better tumor control than monospecific CAR-T cells on heterogeneous models. To overcome tumor heterogeneity, we targeted mesothelin (meso) and Mucin 16 (MUC16), two antigens commonly expressed in solid tumors, using a tandem CAR design.

The concept of optimality dominates contemporary human movement science, with researchers across biomechanics, motor control, and neuroscience routinely explaining observed behaviors as solutions that maximize or minimize objective functions. This paper critiques the pervasive application of optimality principles in human movement science. We argue that optimization frameworks mischaracterize biological systems for several reasons: (1) Evolution produces sufficient rather than optimal adaptations without foresight; (2) Biological systems serve multiple functions simultaneously with context-dependent prioritization; (3) Structure-function relationships co-evolve rather than optimize for fixed targets; (4) The fractal, multiscale nature of physiological signals makes traditional optimization mathematically meaningless-there are no well-defined minima or maxima in fractal landscapes; (5) Optimality models implicitly invoke a homunculus that selects optimization criteria; and (6) The concept is methodologically circular and unfalsifiable, as any behavior can be retroactively modeled as optimal for some function.

Background: Biological age (BA) is increasingly recognized as a valuable alternative to chronological age (CA) for assessing an individual's health and aging status. However, existing models are based on limited clinical parameters and have not thoroughly integrated morbidity and mortality data.
Objective: This study aimed to develop and validate a novel transformer-based model, referred to as the BA - CA gap model, for BA estimation that incorporates morbidity and mortality information to improve predictive accuracy and enhance clinical use in the early identification of the risk of age-related diseases.
