Assessment of organophosphorus (OP) pesticide exposure in non-target organisms rarely involves non-neural molecular targets. Here we performed a 30-d microcosm experiment with Lumbricus terrestris to determine whether the activity of digestive enzymes (phosphatase, β-glucosidase, carboxylesterase and lipase) was sensitive to chlorpyrifos (5 mg kg wet soil). Likewise, acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) activities were measured in the wall muscle and gastrointestinal tissues as indicators of OP exposure. Chlorpyrifos inhibited the acid phosphatase (34% of controls), carboxylesterase (25.6%) and lipase activities (31%) in the gastrointestinal content. However, in the gastrointestinal tissue, only the carboxylesterase and lipase activities were significantly depressed (42-67% carboxylesterase inhibition in the foregut and crop/gizzard, and 15% lipase inhibition in the foregut). Chlorpyrifos inhibited the activity of both cholinesterases in the gastrointestinal tissues, whereas the AChE activity was affected in the wall muscle. These results suggested chlorpyrifos was widely distributed throughout the earthworm body after 30 d of incubation. Interestingly, we found muscle carboxylesterase activity strongly inhibited (92% of control) compared with that detected in the gastrointestinal tissues of the same OP-exposed individuals. This finding was explained by the occurrence of pesticide-resistant esterases in the gastrointestinal tissues, which were evidenced by zymography. Our results suggest that digestive processes of L. terrestris may be altered by chlorpyrifos, as a consequence of the inhibitory action of the insecticide on some digestive enzymes.

While the cancer genome is well-studied, the nongenetic exposome of cancer remains elusive, particularly for regionally prevalent cancers with poor prognosis. Here, by employing a combined knowledge- and data-driven strategy, we profile the chemical exposome of plasma from 53 healthy controls, 14 esophagitis and 101 esophageal squamous cell carcinoma (ESCC) patients, and 46 esophageal tissues across 12 Chinese provinces, integrating inorganic, endogenous, and exogenous chemicals. We first show that components of the ESCC chemical exposome mediate the relationship between ESCC-related dietary/lifestyle factors and clinic health status indicators.

Background: Comorbidities and genetic correlations between gastrointestinal tract diseases and psychiatric disorders have been widely reported, but the underlying intrinsic link between Alzheimer's disease (AD) and inflammatory bowel disease (IBD) is not adequately understood.
Methods: To identify pathogenic cell types of AD and IBD and explore their shared genetic architecture, we developed Pathogenic Cell types and shared Genetic Loci (PCGL) framework, which studied AD and IBD and its two subtypes of ulcerative colitis (UC) and Crohn's disease (CD).
Results: We found that monocytes and CD8 T cells were the enriched pathogenic cell types of AD and IBDs, respectively.

Introduction: Pancreatic adenocarcinoma (PAAD) is a highly aggressive malignancy characterized by a profoundly hypoxic tumor microenvironment, which fosters tumor progression and confers resistance to therapy The oncogenic regulator ID1has been implicated in PAAD malignancy, however, the mechanisms underlying hypoxia-induced stabilization of ID1 and the role of ubiquitin-mediated degradation remain poorly understood. Elucidating these pathways is essential for identifying novel therapeutic targets for PAAD.
Methods: In this study, we examined ID1 expression in PAAD tissues and cell lines using publicly available databases and in vitro models.
