Article Abstract

PEG coated vesicles are important vehicles for the passive targeting of anticancer drugs. With a view to prepare PEG decorated vesicles using co-assembly of block copolymers and lipids, here we investigated the microstructure of aggregates formed in mixtures comprising lipids (l-α-phosphatidylcholine) and block copolymers (Pluronic P123), in the polymer rich regime. DLS and SANS studies show that the structure of the aggregates can be tuned from micelles to rod-like micelles or vesicles by changing the lipid to polymer composition. Rheological studies on gels formed by mixtures of polymer and lipid suggest incorporation of the lipid into the polymer matrix. The encapsulation efficiencies of polymer incorporated liposomes for curcumin and doxorubicin hydrochloride (DOX) are evaluated at different drug to carrier ratios. The pH dependent sustained release of both the drugs from the PEGylated liposomes suggests their application in the development of cost effective formulations for anticancer drug delivery.

Glioblastoma multiforme (GBM) continues to pose a significant challenge in the field of neuro-oncology primarily because of the limited penetration of therapeutics across the blood-brain barrier (BBB) and the presence of immunosuppressive tumor microenvironments. To address these challenges, a HD-PEG@BM biomimetic nanoplatform (hereinafter referred to as HD-P@BM) is developed that cloaks the near-infrared II photosensitizer HD-PEG (HD-P) inside microglial membranes to enable enhanced BBB penetration and tumor-targeted delivery. In this study, it is found that the microglia-derived membranes enhanced the uptake of nanoparticles by both the glioma cells and tumor-associated microglia.

DNA origami-based nanotechnology is a versatile tool for exploring fundamental biological questions and holds significant promise for future biomedical applications. Here, we leverage the optical transparency of the embryonic zebrafish to analyze live embryos injected intravenously with fluorescently labeled wireframe DNA origami nanosheets. Our approach integrated long-term, high-resolution imaging of transgenic live zebrafish embryos with single-cell RNA sequencing to elucidate the effects of oligolysine-polyethylene glycol copolymer (K-PEG) coating on the biodistribution of fluorescence signal in embryos injected with wireframe DNA origami nanosheets.

Collagen VI is an extracellular matrix protein forming complex microfibrillar networks in connective tissues. Specifically, we focused on its role in innate immunity, in particular on cationic sequence motifs from the α3(VI)-chain, which exhibit strong antibacterial properties against both Gram-positive and Gram-negative bacteria in vitro and in vivo. Cytotoxicity assays revealed minimal to no adverse effects, even at concentrations effective against bacterial pathogens.
