Monoamine oxidases (MAOs) play an important role in Alzheimer's disease (AD) pathology. We report comonitoring of MAO activity and amyloid-β (Aβ) plaques dependent on the aging of live mice with AD, using a two-photon fluorescence probe. The probe under the catalytic action of MAO produces a dipolar fluorophore that senses Aβ plaques, a general AD biomarker, enabling us to comonitor the enzyme activity and the progress of AD indicated by Aβ plaques. The results show that the progress of AD has a close correlation with MAO activity, which can be categorized into three stages: slow initiation stage up to three months, an aggressive stage, and a saturation stage from nine months. Histological analysis also reveals elevation of MAO activity around Aβ plaques in aged mice. The close correlation between the MAO activity and AD progress observed by monitoring for the first time prompts us to investigate the enzyme as a potential biomarker of AD.

Abscisic acid (ABA) and calcium respectively work as crucial plant hormones and second signalling molecules in the regulation of fruit development and quality formation, including the sugar content and flavour quality. However, the regulatory mechanisms of fruit sugar accumulation arising from the interaction between ABA and calcium have not yet been fully elucidated. Here, we show that the application of ABA or calcium enhances sugar accumulation in sweet orange (Citrus sinensis) fruit, accompanied by upregulation of the expression level of tonoplast sugar transporter 2 (CsTST2), which mediates the transport of sugars into the vacuole.

Background: Prostate cancer is one of the principal malignancies threatening human health, and the development of castration resistance often constitutes a major cause of treatment failure in its management.
Methods: To elucidate the potential association between programmed death-ligand 1 (PD-L1) and castration resistance in prostate cancer, we analyzed the expression levels of PD-L1 in both primary prostate cancer tissues and castration-resistant prostate cancer (CRPC) specimens as well as in corresponding cell lines by using western blots and immunohistochemistry. Delivering therapeutics across the blood-brain barrier (BBB) remains a major challenge in ischemic stroke therapy. Ischemic stroke induces upregulation of various inflammatory membrane receptors on brain endothelial cells, offering potential entry points for receptor-mediated transcytosis. This study proposes a universal targeting strategy by employing inflammatory pathway antagonists as targeting ligands, which broadens the spectrum of available ligands beyond traditional receptor-binding molecules.

As a dual-purpose medicinal and edible plant, Ocimum species offer significant pharmacological potential through their alkaloid metabolites. This study comprehensively analyzed alkaloid profiles in 10 Ocimum accessions using ultra performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS), integrated with network pharmacology, molecular docking, and transcriptome sequencing to elucidate key pharmacological targets, therapeutic potentials, and biosynthetic pathways. We identified 191 alkaloids categorized into eight classes, with phenolamine and plumerane alkaloids predominating.
