Article Abstract

Fasting glucose and insulin are intermediate traits for type 2 diabetes. Here we explore the role of coding variation on these traits by analysis of variants on the HumanExome BeadChip in 60,564 non-diabetic individuals and in 16,491 T2D cases and 81,877 controls. We identify a novel association of a low-frequency nonsynonymous SNV in GLP1R (A316T; rs10305492; MAF=1.4%) with lower FG (β=-0.09±0.01 mmol l(-1), P=3.4 × 10(-12)), T2D risk (OR[95%CI]=0.86[0.76-0.96], P=0.010), early insulin secretion (β=-0.07±0.035 pmolinsulin mmolglucose(-1), P=0.048), but higher 2-h glucose (β=0.16±0.05 mmol l(-1), P=4.3 × 10(-4)). We identify a gene-based association with FG at G6PC2 (pSKAT=6.8 × 10(-6)) driven by four rare protein-coding SNVs (H177Y, Y207S, R283X and S324P). We identify rs651007 (MAF=20%) in the first intron of ABO at the putative promoter of an antisense lncRNA, associating with higher FG (β=0.02±0.004 mmol l(-1), P=1.3 × 10(-8)). Our approach identifies novel coding variant associations and extends the allelic spectrum of variation underlying diabetes-related quantitative traits and T2D susceptibility.

Dynamics of Conventional Metabolic Indices in Relation to Endometriosis Severity: A Retrospective Analysis.

Int J Gen Med

Department of Gynecology, Zhongshan Hospital, Fudan University, Shanghai, 200035, People's Republic of China.

Objective: This study aims to investigate the association between the dynamics of routine metabolic markers and endometriosis severity.

Methods: A retrospective analysis was conducted on patients diagnosed with endometriosis at Zhongshan Hospital, Xiamen, affiliated with Fudan University. The collected data encompassed demographic details and biochemical indicators related to lipid, hepatobiliary, renal metabolism, and electrolyte balance.

Background: Glucagon-like peptide-1 receptor agonists (GLP-1RAs) are established treatments for obesity. However, it remains inconclusive whether the combination of lifestyle modifications and GLP-1RA interventions can lead to greater weight loss and better control of cardiovascular biomarkers. We aimed to evaluate the efficacy of this combination therapy on weight loss and cardiometabolic markers in adults with overweight or obesity.

Background And Aim: is a significant pathogen in freshwater aquaculture, contributing to high morbidity and mortality in common carp (). Conventional reliance on antibiotics raises concerns about resistance and environmental impact. This study aimed to evaluate the effects of short-term fasting (1 or 2 days) on physiological, oxidative stress, and microbial responses in infected with .

Background: Schizophrenia (SCZ) is a common, chronic, severe mental disorder that is often accompanied by dyslipidemia and linked to decreased life expectancy. The prevalence of dyslipidemia among initial-treatment and drug-naïve (ITDN) patients with SCZ and the correlates influencing its occurrence and severity were determined in this study.

Methods: Demographic and clinical data including blood pressure, blood cell count, renal function, lipid profile, fasting glucose level, and thyroid function were collected from the 668 patients with ITDN SCZ included in this study.

Aim: Sodium-glucose cotransporter 2 (SGLT2) inhibitors consistently demonstrate renal protection against progressive kidney disease. We hypothesised that SGLT2 inhibition reduces blood glucose levels in peri-proximal tubular capillaries by limiting reabsorption from the tubular filtrate, thereby safeguarding the renal microvasculature from hyperglycaemic stress.

Materials And Methods: In anaesthetised streptozotocin-induced type 1 and Otsuka-Long Evans fatty (OLETF) type 2 diabetic rats, we measured the arterial-to-renal venous glucose ratio (RV/A) to evaluate the effects of canagliflozin, a SGLT2 inhibitor.
