: Vericiguat, a soluble guanylate cyclase stimulator, reduces cardiovascular events in patients with heart failure with reduced ejection fraction following clinical deterioration against guideline-directed medical therapy. However, the optimal timing for initiating vericiguat remains unclear. : We retrospectively analyzed heart failure patients with reduced/mild-reduced ejection fraction who received vericiguat between 2021 and 2025 upon optimal guideline-directed medical therapy.

Carbon monoxide (CO) is generally recognized as a toxic gas; however, it has recently been identified as an endogenous gasotransmitter with significant cytoprotective properties. CO modulates key molecular pathways, including anti-inflammatory, anti-apoptotic, antioxidant, and vasodilatory signaling pathways, by targeting heme- and non-heme-containing proteins. These proteins include soluble guanylate cyclase, cytochrome P450 enzymes, MAPKs, and NLRP3.

Background: Endothelial cells (ECs) induce vascular smooth muscle cells (VSMCs) relaxation via nitric oxide (NO), prostacyclin (PGI₂) and hyperpolarizing factors. Recent whole-genomic, single-cell transcriptomic analysis of human vascular cells has revealed angiotypic heterogeneity. However, it remains unknown whether vasorelaxant mediators reiterate this pattern.

Background: Type-II diabetes (DMII) and metabolic syndrome (MBS) increase ventricular arrhythmia and sudden cardiac death risk.
Objectives: Identify mechanisms through which DMII and MBS promote ventricular tachycardia (VT).
Methods: We performed programmed ventricular stimulation (PVS) on leptin receptor mutant (Db/Db) mice with DMII, high fat high sucrose (HFHS)-fed mice with MBS, and cGMP-dependent Protein Kinase 1α (PKG1α) leucine zipper mutant (LZM) mice, which have neither DMII nor MBS but have disrupted PKG1α signaling.

Systemic sclerosis (SSc) is a rare, multisystem autoimmune connective tissue disease characterized by microvascular dysfunction, inflammation, and progressive fibrosis. The pathogenesis of SSc is regulated by both epigenetic and genetic factors. Although the pathogenesis is being explored, treatment options, especially for fibrosis, remain limited.
