Article Abstract

Protein engineering by random circular permutation is an effective tool for tailoring protein topology with potential functional benefits including improved catalytic activity. This method involves covalently connecting the native protein termini with a peptide linker and cleaving a peptide bond elsewhere in the polypeptide sequence. Termini relocation can impact protein ternary and quaternary structure and translate into functional enhancements due to changes in protein conformation and flexibility. As the effects of new termini in specific protein locations are difficult to predict, the preparation of a library constituting all possible permutation sites is an effective search strategy for identifying variants with novel properties.

This report sought to employ multi-modal integration of pre-treatment brain (electroencephalogram, resting-state functional magnetic resonance imaging) and blood (immune and metabolic) biomarkers to facilitate causal inference-based treatment selection by virtue of establishing predictability of remission to multi-stage antidepressant treatment. Data from two stages of pharmacotherapy in the 'Establishing Moderators and Biosignatures of Antidepressant Response for Clinical Care for Depression' (EMBARC) study from participants with both brain and blood biomarkers were included (N = 197). Participants were initially randomized to sertraline or placebo (Stage 1), and depending on clinical response at week-8, their therapy in Stage 2 was either maintained or switched (to sertraline, if a non-responder to placebo, or to bupropion, if a non-responder to sertraline).

Purpose: to evaluate the effect of a novel controlled dynamization device attached to external circular fixators on bone healing of tibial fractures.

Methods: this double-blinded randomized clinical trial evaluated 43 patients over 18 years of age with complex tibial pathologies who were treated without (control) or with a 3D-printed controlled dynamization device attached to external fixators. The devices were installed 60 days after fracture fixation surgery and used for 30 days.
