Rapamycin at high doses (2-10 mg/kg body weight) inhibits mammalian target of rapamycin complex 1 (mTORC1) and protein synthesis in mice. In contrast, low doses of rapamycin (10 μg/kg) increase mTORC1 activity and protein synthesis in skeletal muscle. Similar changes are found with SLF (synthetic ligand for FKBP12, which does not inhibit mTORC1) and in mice with a skeletal muscle-specific FKBP12 deficiency. These interventions also increase Ca(2+) influx to enhance refilling of sarcoplasmic reticulum Ca(2+) stores, slow muscle fatigue, and increase running endurance without negatively impacting cardiac function. FKBP12 deficiency or longer treatments with low dose rapamycin or SLF increase the percentage of type I fibers, further adding to fatigue resistance. We demonstrate that FKBP12 and its ligands impact multiple aspects of muscle function.

Two major protein recycling pathways have emerged as key regulators of enduring forms of synaptic plasticity, such as long-term potentiation (LTP), yet how these pathways are recruited during plasticity is unknown. Phosphatidylinositol-3-phosphate (PI(3)P) is a key regulator of endosomal trafficking and alterations in this lipid have been linked to neurodegeneration. Here, using primary hippocampal neurons, we demonstrate dynamic PI(3)P synthesis during chemical induction of LTP (cLTP), which drives coordinate recruitment of the SNX17-Retriever and SNX27-Retromer pathways to endosomes and synaptic sites.

This study examined the effects of 24R,25-dihydroxyvitamin D (24R,25(OH)D) in estrogen-responsive laryngeal cancer tumorigenesis in vivo, the mechanisms involved, and whether the ability of the tumor cells to produce 24R,25(OH)D locally is estrogen-dependent. Estrogen receptor alpha-66 positive (ER+) UM-SCC-12 cells and ER- UM-SCC-11A cells responded differently to 24R,25(OH)D in vivo; 24R,25(OH)D enhanced tumorigenesis in ER+ tumors but inhibited tumorigenesis in ER- tumors. Treatment with 17β-estradiol (E) for 24 h reduced levels of CYP24A1 protein but increased 24R,25(OH)D production in ER+ cells; treatment with E for 9 min reduced CYP24A1 at 24 h and reduced 24R,25(OH)D production in ER- cells.
