A new method for the construction of tetrahydropyrans derived from readily available 1,2-dioxines containing a tethered hydroxyl moiety is described. The reaction proceeds via a base-catalyzed rearrangement of the 1,2-dioxines to either the isomeric cis or trans gamma-hydroxy enones followed by intramolecular oxa-Michael addition of the tethered hydroxyl group.

Given the urgent need for developing new therapeutic strategies against chronic diseases such as neurodegeneration and cancer, and the evidence of shared biochemical pathways linking Alzheimer's disease to certain types of cancer, we have designed a novel family of multifaceted compounds to target both diseases. For that purpose, isofagomine (a relevant azasugar) - coumarin hybrids were prepared and tested in vitro as potential dual-action molecules. Key structural variations, including different hydroxyl group substitutions and changes in the length of the hydrocarbon linker, had minimal impact on cholinesterase inhibition.

Biosynthetic studies of the platensimycin (PTM), platencin (PTN), and platensilin (PTL) family of natural products have revealed numerous insights into the chemistry and enzymology of diterpenoid biosynthesis. A deeper understanding of the PTM biosynthetic machinery would advance fundamental knowledge in natural product biosynthesis and facilitate future efforts to exploit these compounds as potential leads for biomedical applications. Herein, we report the functional characterization of the gene that encodes the dehydratase responsible for the formation of the C6-C7 enone moiety in PTM, PTN, and PTL.
