Objectives: To investigate the therapeutic mechanism of 2,6-dimethoxy-1,4-benzoquinone (DMQ) for alleviating dextran sulfate sodium (DSS)-induced ulcerative colitis (UC) in mice.

Methods: Eighteen male C57BL/6J mice were equally randomized into control group, DSS group and DMQ treatment group. In DSS and DMQ groups, the mice were treated with DSS in drinking water to induce UC, and received intraperitoneal injections of sterile PBS or DMQ (20 mg/kg) during modeling.

Butyl acetate, a valuable flavor ester, is conventionally synthesized through acid-catalyzed reactions, which suffer from environmental concerns and inefficiencies. This study explores a greener alternative using liquid lipase Novozym 400238 for its enzymatic synthesis. The central composite design (CCD) within response surface methodology (RSM) was employed to assess the reaction parameters, including temperature, substrate molar ratio, enzyme concentration, and hexane content, along with their effects on the conversion rate.

DGKα, also named diacylglycerol kinase alpha, plays an important role in signal transduction, phosphorylating the membrane lipid diacylglycerol (DAG), to phosphatidic acid (PA). Increasing evidence indicates that DGKα-mediated T-cell dysfunction plays a significant role in the development of resistance of the PD-1 blockade. In this article, we report the discovery of compound as a novel, potent, orally available DGKα inhibitor with excellent kinase selectivity, a favorable ADME profile, and robust in vivo antitumor activity in combination with anti-PD-1 therapy.
