This work demonstrates for the first time the possibility of using a novel ionic liquid 3-(2-hydrazinyl-2-oxoethyl)-1-methyl-1H-imidazol-3-ium chloride ([HOMI]Cl) as a derivatization reagent for steroid hormones and their subsequent analysis using ultra-high performance liquid chromatography-quadrupole time-of-flight mass spectrometry in human urine. Derivatization conditions, such as pH, reagent concentration, reaction time and temperature were evaluated and optimized. A comparative study of [HOMI]Cl and hydroxylamine effectiveness revealed a formation of both syn- and anti-forms of the steroid derivatives for the novel [HOMI]Cl reagent similarly to a derivatization with hydroxylamine.

Fatty acid esters of hydroxy fatty acids (FAHFAs) are emerging bioactive lipids in edible oils, but their trace-level presence and matrix complexity hinder accurate analysis. Here, we designed a novel chemoselective probe piperazine-modified silica () that specifically captures FAHFAs while removing almost totally triglycerides in a one-step labeling reaction. The probe enabled single-batch sample pretreatment within 30 min, integrating capture, purification, and signal amplification.

We studied the effect of increasing and decreasing the deuterium level on the mitochondrial membrane potential (MMP), Ca homeostasis, and neuronal death in the rat cerebellar neurons in vitro. It was found that decreasing deuterium concentration to 100 and 50 ppm was accompanied by gradual depression of MMP and a decrease in the level of cytosolic Ca compared to the baseline values at deuterium concentration 150 ppm. Increasing deuterium concentration to 300 and 750 ppm was associated with an increase in MMP and Ca levels.

Scientific Department, Kuban State University, Krasnodar, Russian Federation; Laboratory of Problems of Stable Isotope Spreading in Living Systems, Southern Scientific Center of the Russian Academy of Sciences, Rostov-on-Don, Russian Federation.

The ATXN2 gene is located on chromosome 12q24.1 and encodes the ataxin-2 protein, which is involved in the regulation of RNA metabolism, protein synthesis, and intracellular signaling. The polyQ tract encoding glutamine can expand, which leads to the development of various neurodegenerative diseases.

Psychological studies on close relationships have often overlooked cultural diversity, dynamic processes, and potentially universal principles that shape intimate partnerships. To address the limited generalizability of previous research and advance our understanding of romantic love experiences, mate preferences, and physical attractiveness, we conducted a large-scale cross-cultural survey study on these topics. A total of 404 researchers collected data in 45 languages from April to August 2021, involving 117,293 participants from 175 countries.

The nucleotide sequence determines the structural properties of B-DNA; this fact is considered key in recognizing protein binding sites and changing its activity. Our study has established that, using an angular mathematical model, it is possible to determine areas of similar collective states and bends in the nucleotide sequence that arise even under small torque influences. Calculations were performed for two genes that differ slightly in length ( and fragment of the gene to compare).

Clinical data as well as animal and cell studies indicate that certain antidiabetic drugs, including glucagon-like peptide 1 receptor agonists (GLP-1RAs), exert therapeutic effects in Alzheimer's disease (AD) by modulating amyloid-β peptide (Aβ) metabolism. Meanwhile, the direct interactions between GLP-1RAs and Aβ and their functional consequences remain unexplored. In this study, the interactions between monomeric Aβ40/Aβ42 of GLP-1(7-37) and its several analogs (semaglutide (Sema), liraglutide (Lira), exenatide (Exen)) were studied using biolayer interferometry and surface plasmon resonance spectroscopy.

It is known that a number of neurodegenerative diseases, also called diseases of waiting, are associated with the expansion of the polyQ tract in the first exon of the gene. In the expanded polyQ tract, the probability of occurrence of non-canonical configurations (hairpins, G-quadruplexes, etc.) is significantly higher than in the normal one.

The relevance of the hemodialysis procedure is increasing worldwide due to the growing number of patients suffering from chronic kidney disease. Taking into account the structure of dialysis polymer membranes is an important aspect in their development to achieve the required performance of hemodialyzers. We propose a new mathematical model of mass transfer that allows hollow-fiber membrane structural parameters to be taken into account in simulating the clearance () of hemodialyzers in a way that does not require difficult to achieve close approximation to the exact geometry of the membrane porous structure.

Given the ubiquitous nature of love, numerous theories have been proposed to explain its existence. One such theory refers to love as a commitment device, suggesting that romantic love evolved to foster commitment between partners and enhance their reproductive success. In the present study, we investigated this hypothesis using a large-scale sample of 86,310 individual responses collected across 90 countries.

Thin-film membranes of Pd-Ag and Pd-Cu alloys capable of releasing hydrogen in a wide temperature range have been developed. The surface activation of the membranes with a nanostructured coating made it possible to intensify hydrogen transport through Pd-containing membranes at low temperatures. This effect was achieved by accelerating limiting surface processes by increasing the active area of the membrane.

Background: There is an assumption about the presence of a specific nucleotides sequence in DNA molecule, which contributes to the genesis of open states (OS). In addition, it would be logical to assume that OS zones should form in DNA regions with a large proportion of Adenine-Thymine (AT) pairs, since they contain fewer hydrogen bonds than Guanine- Cytosine (GC) base pairs. However, studies have shown that in areas rich in AT pairs, the probability of open states will not always be higher.

Pulsed electric field (PEF) modes of electrodialysis (ED) are known for their efficiency in mitigating the fouling of ion-exchange membranes. Many authors have also reported the possibility of increasing the mass transfer/desalination rate and reducing energy costs. In the literature, such possibilities were theoretically studied using 1D modeling, which, however, did not consider the effect of electroconvection.

Trinucleotide repeats are the cause of many neurodegenerative diseases that are currently incurable. In this regard, the question of the causes of occurrence and methods of prevention or treatment of diseases caused by the expansion of repeats in the CAG tract of the gene remains relevant. Previously, it was shown that the frequency of occurrence of additional OS (open states) zones increases with increasing length of the CAG tract, and the value inverse to the frequency correlates with the age of disease onset.

Quantification of cancer biomarkers in urine using volatilomic approach.

Heliyon

October 2024

Shenzhen MSU-BIT University, International University, Park Road, 1, Shenzhen, Longgang District, Guangdong Province, 518172, PR China.

Urine analysis is an attractive approach for non-invasive cancer diagnostics. In this study, a procedure for the determination of volatile organic compounds (VOCs) in human urine (acetone, acetonitrile, dimethylsulfide, dimethyl disulfide, dimethyl trisulfide, hexane, benzene, toluene, 2-butanone, 2-pentanone, pentanal) has been described including sample preparation using preconcentration of analytes in sorbent tubes followed by gas chromatography with mass spectrometry (GC-MS). Fractional factorial design and constrained surfaces design were used to optimize preconcentration of VOCs in sorbent tubes.
