Potato (Solanum tuberosum L.) yield is highly sensitive to drought stress, yet robust phenotyping methods for drought tolerance remain scarce. To address this challenge, we present a rapid, high-throughput hydroponic assay designed as an efficient pre-screening tool for evaluating potato cultivars and CRISPR-edited lines. This protocol serves as a foundational screen, enabling researchers to identify the most promising genotypes before committing resources to more extensive soil or field trials. The system uses repurposed pipette tip boxes as low-cost, scalable hydroponic units, making the method highly accessible. Over a five-week period, plantlets are subjected to controlled 24-hr osmotic stress with polyethylene glycol (PEG-6000) and then assessed for resilience based on biomass, photosynthetic measurements, and visual recovery. This resource-efficient assay provides a controlled environment to minimize experimental noise and has been successfully applied to characterize CRISPR-edited potato lines. By providing a reproducible platform for initial evaluation, this protocol accelerates the selection pipeline for developing robust potato varieties for a changing climate. © 2025 The Author(s). ConspectusMolecular photochemistry, by harnessing the excited states of organic molecules, provides a platform fundamentally distinct from thermochemistry for generating reactive open-shell or spin-active species under mild conditions. Among its diverse applications, the resurgence of the Minisci-type reaction, a transformation historically reliant on thermally initiated radical conditions, has been fueled by modern photochemical strategies with improved efficiency and selectivity. Consequently, the photochemical Minisci-type reaction ranks among the most enabling methods for C()-H functionalizations of heteroarenes, which are of particular significance in medicinal chemistry for the rapid diversification of bioactive scaffolds.

Aims/hypothesis: Alpha cell dysregulation is an integral part of type 2 diabetes pathophysiology, increasing fasting as well as postprandial glucose concentrations. Alpha cell dysregulation occurs in tandem with the development of insulin resistance and changes in beta cell function. Our aim was to investigate, using mathematical modelling, the role of alpha cell dysregulation in beta cell compensatory insulin secretion and subsequent failure in the progression from normoglycaemia to type 2 diabetes defined by ADA criteria.

Despite the widespread utility of transition metal-catalyzed cross-couplings in organic synthesis, the coupling of unactivated alkyl electrophiles remains challenging due to sluggish oxidative addition and competing side reactions. Here, we describe a general and practical copper-catalyzed radical deoxyalkynylation of α-unfunctionalized alcohols through a synergistic combination of Barton-McCombie deoxygenation and copper-catalyzed radical cross-coupling. Key to the success of this method lies in not only the development of rigid anionic multiple N,N,N-ligand to exert remarkable selectivity of highly reactive unactivated alkyl radicals, but also the selection of one suitable oxidant to suppress Glaser homocoupling and other side products.
