Article Abstract

Geraniin is a plant natural product that has been testified to perfect osteoporosis (OP). However, Geraniin's impacts on osteoblast autophagy and its latent molecular mechanisms remain uncertain. In this study, Geraniin's impacts on autophagy in osteoblasts were figured out and its molecular mechanism was uncovered. We established in vivo and in vitro models of OP using glucocorticoids and administered Geraniin. The results manifested that Geraniin dose-dependently promoted the phosphorylation of PI3K/Akt/mTOR in OP rats and osteoblasts, increased the bone mineral density, bone surface area/bone volume, Trabecular number and trabecular bone thickness of rat femurs, enhanced alkaline phosphatase activity in OP rats and osteoblasts, elevated osteogenesis-linked proteins RUNX2, OSX and OCN, facilitated autophagosome formation in osteoblasts, elevated Beclin-1 and ACTG5 expression levels and LC3II/LC3I ratio and downregulated p62. However, these effects of Geraniin were attenuated by PI3K/Akt/mTOR inhibitors. All in all, the results of this study indicate Geraniin can perfect glucocorticoid-induced OP by motivating autophagy in osteoblasts through PI3K/Akt/mTOR cascade, which offers strong data support for the future development of Geraniin as a drug for OP cure.

Background: To elucidate the therapeutic effects and underlying mechanisms of palmatine, a principal alkaloid derived from Coptis chinensis, on neuroinflammation in ischemic stroke rat models induced by middle cerebral artery occlusion (MCAO).

Methods: Initially, qPCR was employed to assess the impact of neurotrophic factors secreted by SH-SY5Y neuroblastoma cells on the phenotypes of BV2 cells. Alterations in sphingolipid profiles within neuronal supernatants were characterized using liquid chromatography-tandem mass spectrometry, and molecular docking studies were conducted to investigate the interaction of palmatine with key enzymes involved in sphingolipid metabolism.

Primary cutaneous T-cell lymphoma (CTCL) manifests as a distinct variant of T-cell non-Hodgkin's lymphoma, predominantly impacting skin tissues and constituting approximately 75%-80% of cutaneous lymphoma cases, exhibiting diverse clinical presentations. Sezary syndrome (SS) is a rare subtype of CTCL. The therapeutic approach of SS frequently incorporates multiple chemotherapeutic compounds, encompassing specific histone deacetylase inhibitor agents.

Acute myeloid leukemia (AML) is a hematologic malignancy with a high mortality rate and poor prognosis, largely attributed to the emergence of chemotherapy resistance. Cytarabine (Ara-C), the cornerstone chemotherapeutic agent for AML, faces significant challenges due to the development of resistance, creating an urgent need for novel therapeutic strategies. Pyroptosis as a new form of programmed cell death has emerged as a potential therapeutic target in tumor treatment.

Background: Premature ovarian insufficiency (POI) is a significant health issue for women under 40, characterized by declining ovarian function, hormonal imbalance, and various long-term health consequences. Current hormone replacement therapy (HRT) has limitations and potential risks. Thus, there is a need for safer and more effective therapeutic alternatives.

Computational Structural Biology Section, Frederick National Laboratory for Cancer Research MD 21702 USA

Here, we shed physico-chemical light on major kinase signal transduction cascades in cell proliferation in the Ras network, MAPK and PI3K/AKT/mTOR. The cascades respond to external stimuli. The kinases are allosterically activated and relay the signal, leading to cell growth and division.
