Unlabelled: An investigation of the mangrove-derived fungus sp. DM27 led to the isolation of 19 new compounds, including three pairs of piperidinone enantiomers ( ±)-, ( ±)-, and ( ±)-, two pairs of pyrrolidinone enantiomers ( ±)- and ( ±)-, and nine pyrrolidine derivatives -. The structures of - were elucidated through NMR and HRESIMS analysis, coupled with experimental and calculated ECD spectroscopy and the modified Mosher method. Quantitative real time PCR and Western bolt analyses revealed that blocked EMT in TGF-β1-treated HK-2 cells and suppressed fibroblast activation in TGF-β1-stimulated NIH-3T3 cells. Molecular simulations demonstrated that compound could dock ADAM17, showing a high negative binding affinity. Additionally, the overexpression of ADAM17 by lentiviral infection triggered renal tubular EMT, while compound suppressed this process. Overall, our research suggests that pyrrolidine derivatives may be potential therapeutic agents for the treatment of fibrotic kidney disease.

Direct-acting antivirals (DAAs) have transformed hepatitis C virus (HCV) treatment in Australia since their inclusion on the Pharmaceutical Benefits Scheme (PBS) in 2016. Treatment has shifted from genotype-specific to pan-genotypic regimens, with glecaprevir/pibrentasvir and sofosbuvir/velpatasvir now recommended in clinical guidelines. This study examined trends in DAA dispensing in light of evolving treatment regimens.

Background And Aims: Sofosbuvir (SOF) plus daclatasvir (DCV) is a primary chronic hepatitis C virus (HCV) treatment in low- and middle-income countries. WHO guidelines recommend a half-adult dose for children (14-25 kg) based on pharmacokinetic modelling, requiring clinical validation. We evaluated the pharmacokinetics, safety, efficacy and acceptability of DCV (30 mg) and SOF (200 mg) in children weighing 14 to < 17 kg and 17-35 kg.

The first synthesis of polyhydroxylated 3-methylindolizidines is reported. A straightforward domino butenylmagnesium bromide addition/Cope-House reaction to carbohydrate-derived nitrones afforded efficiently the methyl pyrrolidine moiety with good stereoselectivity, which can be reversed by use of Lewis acids. A subsequent one-pot deprotection/deoxygenation/reductive amination step furnished the desired bicyclic architecture, allowing us to afford the desired final products in 3-4 steps from the starting nitrones and 18-30% overall yields.

Despite the various beneficial properties of cannabidiol (CBD), such as antioxidant, anti-inflammatory, analgesic, antidepressant, and anxiolytic activities, its clinical utility is limited due to its notably low bioavailability (BA). To address this issue, we developed an amorphous CBD powder formulation using solvent shift method, which only uses materials approved for food-grade applications. In a pharmacokinetic study in male Sprague-Dawley rats, we orally administered 10 mg/kg of CBD isolate powder with a crystalline structure and our developed amorphous CBD powder formulation.
