Cell membrane vesicles (CMVs) have been extensively used as delivery vehicles for a variety of cargos, which are generally prepared via membrane extrusion. Extruded CMVs are not necessarily to have the outer membrane facing outward due to the randomness of membrane wrapping. Nanoparticles have been used to serve as cores to direct the membrane orientation of CMVs; nevertheless, there is a lack of methods to efficiently sort coreless CMVs of desired orientations. Herein, we utilized a group of functional DNA probes to reveal the random distribution of membrane orientations of coreless CMVs after extrusion, producing either right-side-out vesicles (RSVs) or inside-out vesicles (ISVs). More importantly, DNA probes that protrude out from the outer membrane can serve as handles for efficiently sorting out RSVs from ISVs to produce vesicles with a dominant right-side-out orientation. We investigated three methods to enrich RSVs, including strand displacement reaction, photo cleavage (PC), and enzymatic cleavage. Among them, PC exhibits the highest enrichment efficiency (∼93%) and RSVs purity (∼85.4%), which therefore is recommended for future applications. This work revealed the mixed orientations of coreless CMVs and provided a technical platform to efficiently enrich CMVs of wanted membrane orientations that shall be useful toward a vast array of biomedical applications.

An increasing number of epigenome-wide association studies report tobacco smoking-associated DNA methylation levels. However, comprehensive replication studies remain scarce, particularly in placenta, despite their crucial interest in such a large-scale context. Using DNA methylation data from the EPIC array of 341 new placentas (85 smokers, 219 non-smokers, and 37 former smokers) from the EDEN cohort, we used a candidate approach to replicate maternal smoking-associated CpGs and regions previously identified using the 450K array, and an exploratory approach to discover new associations within EPIC-specific CpGs.

A series of molecular logic gates with multiple biocomputing capabilities have been successfully fabricated by using four antibiotic residues [tetracycline (TET), chloramphenicol (CHL), kanamycin (KAN), and streptomycin (STR)] as inputs. In standard short-read whole-exome sequencing (WES), capture probes are typically designed to target the protein-coding regions (CDS), and regions outside the exons-except for adjacent intronic sequences-are rarely sequenced. Although the majority of known pathogenic variants reside within the CDS as nonsynonymous variants, some disease-causing variants are located in regions that are difficult to detect by WES alone, such as deep intronic variants and structural variants, often requiring whole-genome sequencing (WGS) for detection. Moreover, WES has limitations in reliably identifying pathogenic variants within mitochondrial DNA or repetitive regions.
