Cherry-red tobacco has aroused significant interest in both the academic and industrial fields due to its unique aroma style. In order to analyze the aroma characteristics of Cherry-red tobacco and the intrinsic aroma substance basis, the aroma compounds were analyzed by gas chromatography-mass spectrometry (GC-MS), the aroma characteristics were verified by sensory evaluation, and a correlation analysis was further conducted. The sensory evaluation suggested that Cherry-red tobacco was significantly richer in aroma, containing a total of 11 aromas, among which nutty, bean, herbal, and spicy were the characteristic aromas. The analysis of aroma compounds indicated that there were 88 differentiating aroma compounds between Cherry-red tobacco and Yunyan 87. The correlation analysis revealed that the glutinous sweet aroma of Cherry-red tobacco was strongly correlated with phenols, aromatic hydrocarbons, and aldehydes, while the fresh sweet aroma of Yunyan 87 was strongly correlated to ketones and nitriles. There were 24 characteristic aroma substances with strong correlations with the glutinous sweet aroma of Cherry-red tobacco. Furthermore, based on the odor and OAV analysis results, 2-vinylpyridine, 2,6-dimethylpyridine, scopoletin, 4-methylphenol, and eugenol were the key aroma compounds that highlighted the characteristic aroma of Cherry-red tobacco.

Common flue-cured tobacco (Nicotiana tabacum L.) primarily accumulates nicotine, and its flue-cured leaves exhibit a lemon appearance. In contrast, a spontaneous cherry-red variant (CR60) primarily accumulates nornicotine, accompanied by distinctive red dapples on the cured leaves.

Eleven consecutive N'-n-acylnornicotines from cherry-red tobacco were structurally elucidated and quantitively analyzed using chromatography and mass spectrometry. All of these N'-n-acylnornicotines are first reported in cherry-red tobacco, whereas N'-propionylnornicotine, N'-n-valerylnornicotine, N'-n-nonanoylnornicotine and N'-n-undecanoylnornicotine are first reported in natural products. The concentration distribution of the identified N'-n-acylnornicotines was studied and it was found that N'-n-octanoylnornicotine and N'-n-hexanoylnornicotine showed the highest concentration, accounting for 94% of the detected N'-n-acylnornicotines.

The genotype CR60 is a spontaneous Cherry Red variant (containing granular red dapples on flue-cured leaves) of the Yunyan 87 (Y87) tobacco; it accumulates higher concentration of iron (Fe) in leaves than Y87, but the physiological differences between them remain largely unknown. We investigated the physiological and molecular mechanisms of CR60 in response to Fe deficiency under hydroponic conditions. Our results showed no significant phenotypic difference between Y87 and CR60 at optimal (40 μM) and high Fe (160 and 320 μM) concentrations.

"Cherry-red" tobacco is the superior variant of tobacco, appearing with the apperance of red dapples on cured leaves due to the demethylation of nicotine to nornicotine during maturation and curing. Fungi are known to have the capacity to convert nicotine to nornicotine. However, an endophytic fungal community of "cherry-red" tobacco has never been reported to our best knowledge.
