Article Abstract

Central nervous system (CNS) resident memory CD8 T cells (T) that express IFN-γ contribute to neurodegenerative processes, including synapse loss, leading to memory impairment. Here, we show that CCR2 signaling in CD8 T that persist within the hippocampus after recovery from CNS infection with West Nile virus (WNV) significantly prevents the development of memory impairments. Using CCR2-deficient mice, we determined that CCR2 expression is not essential for CNS T cell recruitment or virologic control during acute WNV infection. However, transcriptomic analyses of forebrain CCR2 versus CCR2 CD8 T during WNV recovery reveal that CCR2 signaling significantly regulates hippocampal CD8 T phenotype and function via extrinsic and intrinsic effects, limiting expression of CD103, granzyme A and IFN-γ, respectively, and increasing the percentages of virus-specific CD8 T cells. Consistent with this, WNV-recovered Cd8aCcr2 mice exhibit decreased recognition memory. Overall, these data implicate CCR2 signaling in the regulation of CD8 T phenotype, including antiviral specificity and IFN-γ expression, highlighing a neuroprotective role for CCR2 in limiting CD8 T cell-mediated neuroinflammation and cognitive deficits, providing insights into potential therapeutic targets for CNS infections.

CX3CR1 modulates acute disc herniation-induced pain via regulating local inflammation and spinal microglia activation.

Osteoarthritis Cartilage

Department of Orthopaedic Surgery, University of Virginia, Charlottesville, VA 22908, USA; Department of Biomedical Engineering, University of Virginia, Charlottesville, VA 22904, USA. Electronic address:

Objective: Inflammation is a key driver of disc herniation, a major cause of back pain and disability. Heterogeneous macrophages infiltrated at disc hernia sites, yet their role in disease pathology and pain remains unclear. This study investigates the role of CX3CR1⁺ macrophages and microglia in local inflammation and pain using transgenic mouse models and surgically induced disc herniation model.

M1-BMDMs with Wnt5a deletion attenuate liver fibrosis by suppression of Wnt5a/Frizzled 2 axis in hepatic progenitors.

Cell Biosci

August 2025

Institute of Liver Diseases, Key Laboratory of Liver and Kidney Disease of the Ministry of Education, Clinical Key Laboratory of TCM of Shanghai, Shuguang Hospital, Shanghai University of Traditional Chinese Medicine (TCM), Shanghai University of TCM, 528, Zhangheng Road, Pudong District, Shanghai,

Background: Bone marrow-derived macrophages (BMDMs) regulate hepatic progenitor cells (HPCs) differentiation, potentially via the Wnt signaling pathway. While M1-polarized BMDMs (M1-BMDMs) exert anti-fibrotic effects in the liver, Wnt5a is implicated in fibrosis progression. The specific influence of Wnt5a levels within M1-BMDMs on HPCs fate and cirrhosis development remains unclear.

Sepsis-induced myocardial injury (SIMI) significantly contributes to sepsis-related mortality, yet effective therapies remain limited. This study investigated the cardioprotective potential of isopropyl 3-(3,4-dihydroxyphenyl)-2-hydroxypropanoate (IDHP), a bioactive metabolite from , focusing on its mechanism via the GAS6/Axl signaling axis in lipopolysaccharide (LPS)-induced myocardial injury. Using an in vitro HL-1 cardiomyocyte model, IDHP's cytotoxicity was assessed (0-20 μM).

Infiltration of immune cells into the heart plays a crucial role in the transition from adaptive hypertrophy to heart failure (HF) following chronic pressure overload. However, the key factors in myeloid cells that regulate this process are still not well defined. Here, we studied the functional role of S100A8/A9 in myeloid cells during this transition.
