Article Abstract

Exploring three-dimensional chemical space is an important research objective of organic synthetic chemistry. Oxidative dearomatization (ODA) is one of the most important and powerful tools for realizing this goal, because it changes and removes aromatic structures from aromatic compounds to increase levels of saturation and stereoisomerism by direct addition reactions between functional groups with aromatic cores under oxidative conditions. As a hot topic in indole chemistry, the synthetic value of the oxidative dearomatization of indoles has been well recognized and has witnessed rapid development recently, since it could provide convenient and unprecedented access to fabricate high-value-added three-dimensional oxindole skeletons, such as C-quaternary indolones, polycycloindolones and spiroindolones, and be widely applied to the total synthesis of these oxindole alkaloids. Therefore, this article provides a review of recent developments in oxidative dearomatization involving the C-H bonds of indoles. In this article, the features and mechanisms of different types of ODA reactions of indoles are summarized and represented, and asymmetric synthesis methods and their applications are illustrated with examples, and future development trends in this field are predicted at the end.

The enantioselective [4 + 4] cycloaddition for the construction of cyclooctanoids is a challenging transformation in organic chemistry. Herein, we present the first organocatalytic enantioselective [4 + 4] cycloaddition of furan ortho-quinodimethanes, activated by dearomatization of the heteroaromatic compound, which thereby allows for the cycloaddition with dienes. The [4 + 4] cycloaddition is catalyzed by a quinine-derived primary amine in combination with a chiral phosphoric acid and a carboxylic acid affording cyclooctanoids isolated as a single diastereoisomer in good yields and with up to 94% ee.

The Diels-Alder reaction of benzenes remains a significant synthetic challenge, owing to their highly stabilized aromatic cores. In this work, the dearomatization agent {WTp(NO)(PMe)} is used to promote Diels-Alder reactions of dihapto-coordinated (η) benzenes with alkynes. The resulting η-barrelene complexes can be oxidized to liberate intact barrelenes.

We report an asymmetric total synthesis of (-)-neocucurbol C, a diterpene natural product possessing a unique and complex 6/6/5/5/6 polycyclic skeleton and nine stereocenters. Pattern-recognition analysis led us to the chiral pool molecule (+)-nootkatone as the starting material, already containing the AB ring system and two key stereocenters encoded by the target molecule. The extra isopropenyl group of (+)-nootkatone was removed by Kwon's hydrodealkenylative bond fragmentation.

The development of novel anticancer agents is crucial to the ongoing effort to combat cancer. In this study, six-membered phosphorus heterocycles (phosphinanes) were designed, synthesized, and evaluated as potential antiproliferative agents. A series of novel phosphinane derivatives was obtained through structural modifications of 1-phenylphosphinane 1-oxide and 1-phenylphosphinan-4-one 1-oxide, achieving high overall yields.

The reductive dearomatization reaction of N-alkylindoles represents an expedient approach to the generation of N-alkylindolines that are primed for elaboration into more complex target molecules but difficult to access in synthetically acceptable yields directly from (NH)-indolines via their N-alkylation. However, its application to obtain N-alkylindolines bearing reducible functional groups remains a more challenging and long-standing problem due to functional group compatibility issues. In this context, we have achieved a dearomative, chemoselective reduction of epoxide-tethered indoles promoted by NaBHCN/AcOH to obtain hitherto unreported trans-benzylic epoxide-tethered indolines.
