Cancer is considered the uncontrolled growth and spread of cells into neighboring tissues, a process governed at the molecular level by many different factors, including abnormalities in the protein family's death-associated kinase (DAPK). DAPK2 is a member of the DAPK protein family, which plays essential roles in several cellular processes. DAPK2 acts as a tumor suppressor, interacting with several proteins, such as TNF, IFN, etc. during apoptosis and autophagy. Expression of DAPK2 causes changes in the structure of the cell, ultimately leading to cell death by apoptosis. In this essay, studies are obtained from Scopus, PubMed, and the Web of Science. According to these investigations, DAPK2 activates autophagy by interacting with AMPK, mTORC1, and p73. Furthermore, DAPK2 induces apoptosis pathway via interacting with the p73 family and JNK. In general, due to the vital role of DAPK2 in cell physiology and its effect on various factors and signaling pathways, it can be a potent target in the treatment of various cancers, including gastric, ovarian, breast, and other prominent cancers.

Death-associated protein kinase 1 (DAPK1) is a Ca/calmodulin-regulated serine/threonine kinase that orchestrates a wide array of cellular activities. It is intricately regulated through multiple mechanisms, including intramolecular signaling and interactions with other proteins, such as kinases and phosphatases. DAPK1 plays a pivotal role in regulating various biological processes, including apoptosis and autophagy, and is implicated in pathogenesis of several disorders, such as cancer, stroke and brain damage, neurodegenerative and within their kinase domains.

The epidermal growth factor receptor tyrosine kinase inhibitor (EGFR-TKI) resistance is often linked to tumor metastasis, making control of metastasis crucial. Here, we identified a critical signaling hub responsible for cancer metastasis and resistance development: mitochondrial cristae remodeling and metabolic reprogramming, using an anoikis-resistant cell model and a mouse tail vein metastasis model. EGFR-TKI-resistant cells exhibited stronger anoikis resistance (AR) and mitochondrial metabolism compared with sensitive cells, making them more prone to metastasis.

Collective cell migration is seen in various biological processes spanning embryonic development, organogenesis, wound healing and, unfortunately, cancer metastasis. Here, we have examined the role of the evolutionarily conserved Target of Rapamycin signalling (TOR) in mediating collective cell movement employing the model of migrating border cells (BCs) in Drosophila oogenesis. Although TOR signalling is classically linked to cell growth, cell proliferation and metabolism, here we demonstrate that TOR complex 1 (TORC1) regulates efficient group cell movement of BCs.

Many environmental pollutants are known to exhibit neurotoxic effects; however, the initiating molecular events and their associated adverse outcome pathways (AOPs) remain largely unclear. This study investigated the role of circRNA266 and its target miRNA in Triclosan (TCS)-induced neurodevelopmental toxicity in zebrafish (Danio rerio) based on candidates circRNAs identified via high-throughput sequencing. Toxicological effects were characterized at multiple levels, including phenotype alterations, motor behavior, neural damage, and the expression of marker genes.

However, the role of DAPK1 in the pathogenesis of amyotrophic lateral sclerosis (ALS) remains unclear.
