Article Abstract

Aiming to decrease the recurrence of tumors and achieve patient satisfaction, the elicitation of immunotherapy and its integrated synergistic employment is a bright new direction in oncotherapy, yet an emergently challenging task. In particular, tumor-associated macrophage (TAM) regulation though light-induced photodynamic and photothermal therapy (PDT and PTT) is regarded as a powerful approach, which focuses on the systemic immune system instead of the tumor itself. Herein, this study reports an acceptor-donor-acceptor (A-D-A) aggregation-induced emission luminogen (AIEgen), named TPA-2CN, which was applied as a photosensitizer (PS) and photothermal agent (PTA). Attributed to its A-D-A structure and AIE properties, TPA-2CN exhibits a high molar absorption coefficient and acts as a perfect template in regulating radiative and nonradiative transitions, which mainly utilize excited energy. The generation of type I reactive oxygen promoted its application in hypoxic tumor sites and the combination of hyperpyrexia forcefully induces macrophages to polarize towards the immune response M1 phenotype. In and , the successful reversion and reprogramming of the immune microenvironment was impressively proved. This method optimally concentrated immune therapy, PDT and PTT as one and exhibited excellent synergistic therapeutic effects with good biosafety.

We report two A-D-A'-D-A type non-fullerene acceptors, CS-3 and CS-4, with twisted backbones and low crystallinity. Despite these structural drawbacks, CS-3-based photomultiplying organic photodetectors exhibit an external quantum efficiency (EQE) of 5600% at 800 nm and detectivity exceeding 10 Jones. This work highlights the potential of non-planar molecular designs for achieving high-gain, broadband, near-infrared detection beyond conventional structure-performance expectations.

Organic photosensitizers: from molecular design to phototheranostics.

Photodynamic therapy (PDT) has emerged as a highly promising approach for tumor treatment, owing to its remarkable spatiotemporal precision and non-invasive characteristics. Nevertheless, the clinical translation of conventional organic photosensitizers remains constrained by inherent limitations, including a low photosensitization effect, limited reactive oxygen species (ROS) production in a hypoxic tumor microenvironment (TME), restricted tissue penetration depth, and inefficient tumor-targeting. To address these challenges, this review examines molecular engineering strategies through rational structure design, focusing on five critical aspects: (i) to promote the intersystem crossing (ISC) process by introducing heavy atoms, designing photosensitizers with a twisted conformation structure or polymerization for amplified ROS generation; (ii) to conquer tumor hypoxia construction of type I photosensitizers, fractional photosensitizers and other radical-generating photosensitizers; (iii) to excite with near-infrared light constructing a D-A structure, fabricating -aggregates, or utilizing two-photon excitation to improve the penetration depth; (iv) to target tumor tissues through conjugating photosensitizers with tumor-specific ligands or gene-encoded fragments to achieve tumor-targeted therapy; and (v) to reduce the off-target effect designing TME-activatable photosensitizers.

We develop a unique trident-like multi-cationic photosensitizer (PS) with an acceptor-donor-acceptor (A-D-A) structure to enhance photodynamic therapy (PDT). The design minimizes PS leakage from membrane camouflaged nanoparticles (<1% in 24 h) hydrophilic anchoring, while boosting ROS generation (13.8-fold higher O than Rose Bengal) through excited state optimization, thus significantly improving tumor-targeted PDT efficiency.
