Proteolysis-targeting chimeras (PROTACs) that engage two biological targets at once are a promising technology in degrading clinically relevant protein targets. Since factors that influence the biological activities of PROTACs are more complex than those of a small molecule drug, we explored a combination of computational chemistry and deep learning strategies to forecast PROTAC activity and enable automated design. A new method named PROTACable was developed for the de novo design of PROTACs, which includes a robust 3-D modeling workflow to model PROTAC ternary complexes using a library of E3 ligase and linker and an SE(3)-equivariant graph transformer network to predict the activity of newly designed PROTACs. PROTACable is available at https://github.com/giaguaro/PROTACable/.

Background: Immigrants continue to face challenges after entering the labor market and remain overrepresented in '3-D jobs' (dirty, difficult, degrading). This study aims to investigate the differences in occupational injury due to accidents (OIA) among immigrants compared to native-born workers in Sweden, and to examine the role of migrant-specific and work factors in these differences.
Methods: This repeated cross-sectional study used nationwide registers including all gainfully employed individuals in 2004-2020 (average annual sample 4.

Three generations of EGFR tyrosine kinase inhibitors (EGFR-TKIs) have shown clinical efficacy in nonsmall cell lung cancer (NSCLC), but acquired resistance mutations─especially the -EGFR─remain a major challenge. Here, we report the identification of a series of pyrrolo[2,3-]pyrimidine derivatives that inhibit C797S-mediated EGFR triple mutants. Among them, compound shows subnanomolar IC values against Ba/F3 EGFR and Ba/F3 EGFR, while sparing wild-type EGFR.

Chronic wounds pose a growing global health challenge. Accurate assessment is essential for monitoring healing, yet traditional two-dimensional methods lack volumetric analysis. Emerging three-dimensional imaging technologies offer enhanced precision, but their clinical validation and prognostic utility remain unclear.
