Article Abstract

This paper aims to provide the details for making affordable single and multichannel liquid aspirators for wet labs. A liquid aspirator is a basic laboratory device that can cost several hundred Euros. We present a < €25 3D print solution that performs equally well in daily lab routines and is compatible with various vacuum sources, including an aquarium pump or household vacuum cleaner. Presently, commercial aspirators cost more than a decent entry-level 3D printer capable of producing all the parts listed in this manuscript. The models were designed with Tinkercad, with easy printing and minimal support in mind. The versatility and the ultra-low-cost solution we presented could ease the daily workflow of researchers in various research fields. Furthermore, it is valuable to high school or undergraduate student labs and community wet labs for science enthusiasts, where funding is generally limited.

The KEAP1-NRF2 pathway regulates cytoprotective responses to endogenous and exogenous stresses induced by reactive oxygen species (ROS) and electrophiles. To maintain oxidative homeostasis, cancer cells increase the transcription of antioxidant genes by acquiring either stabilizing mutations in NRF2 or inactivating mutations in its negative regulator, KEAP1. NR0B1 is a recently identified NRF2 target that supports anchorage-independent growth in KEAP1-mutant non-small cell lung cancer (NSCLC) cells.

The Amphibian Metamorphosis Assay (AMA) was established as a standardized guideline test with the primary purpose of evaluating test materials for potential endocrine activity in the Hypothalamus-Pituitary-Thyroid axis of developing African clawed frogs (Xenopus laevis). The Extended Amphibian Metamorphosis Assay (EAMA) is similar to the AMA with a key difference in that the EAMA terminates at a fixed developmental stage (ie Nieuwkoop and Faber (NF) stage 62) rather than at a fixed exposure time of the AMA (ie, 21-days). This paper summarizes the control data at NF stage 62 for a total of seventeen EAMAs conducted at two different laboratories.

This protocol set focuses on the preparation of the BD FACSAria II/III/Fusion, a cuvette-based cell sorting system commonly found in shared resource settings, to sort submicron samples, including but not limited to virus-like particles (VLPs) and bacteria. This is meant to serve as a proven workflow for staff in general shared resource laboratories (SRL) and individual labs. It is also useful for labs purchasing cuvette-based sorters with similar fluidic paths to the FACSAria Fusion from BD Biosciences, such as the BD FACSSymphony S6 and BD FACSDiscover S8, as well as for specialized SRLs that will need to move away from Influx and MoFlo platforms that are approaching end of life.
