The epithelial-immune microenvironment (EIME) of epithelial tissues has five common elements: (1) microbial flora, (2) barrier, (3) epithelial cells, (4) immune cells, and (5) peripheral nerve endings. EIME provides both constant defense and situation-specific protective responses through three-layered mechanisms comprising barriers, innate immunity, and acquired immunity. The skin is one of the largest organs in the host defense system. The interactions between the five EIME elements of the skin protect against external dangers from the environment. This dysregulation can result in the generation of inflammatory loops in chronic inflammatory skin diseases. Here, we propose an understanding of EIME in chronic skin diseases, such as atopic dermatitis, psoriasis, systemic lupus erythematosus, alopecia areata, and acne vulgaris. We discuss the current treatment strategies targeting their inflammatory loops and propose possible therapeutic targets in the future.

Sepsis is a complex systemic disease in which systemic toxicity-arising from inflammation-immune dysregulation, oxidative stress, programmed cell death (apoptosis, pyroptosis, ferroptosis), and metabolic reprogramming-drives multi-organ injury. The aim of this review was to synthesize how signaling pathways evolve within and between key organs (lungs, liver, kidneys, heart) and to evaluate whether multi-omics integration and network modeling can identify critical toxic nodes and predict disease progression. We conducted a narrative review of English-language mechanistic studies published between 2015 and 2025 in PubMed, Web of Science, and Scopus, supplemented by bibliography screening, while excluding case reports, conference abstracts, and non-mechanistic work.

Bowel sonography comprises the study of inflammatory bowel disease (IBD) and other inflammatory and neoplastic bowel conditions associated with gross morphologic change. For the most common application, IBD, it is well established that gray-scale US and color Doppler US imaging (CDI) allow excellent assessment of the status of the bowel wall, blood flow to the bowel and mesentery, and the presence or absence of perienteric inflammatory fat. Although these techniques provide effective and accurate methods to assess the bowel, they are not always comprehensive.

Advancements in AI-powered systems medicine have revolutionized biomarker discovery through emergent and explainable features. By use of complex network dynamics and graph-based machine learning, we identified critical determinants of lineage-specific plasticity across the single-cell transcriptomics of pediatric high-grade glioma (pHGGs) subtypes: IDHWT glioblastoma and K27M-altered diffuse midline glioma. Our study identified network interactions regulating glioma morphogenesis via the tumor-immune microenvironment, including neurodevelopmental programs, calcium dynamics, iron metabolism, metabolic reprogramming, and feedback loops between MAPK/ERK and WNT signaling.
