Intestinal organoid technology has revolutionized our approach to cell culture due in part to their three-dimensional structures being more like the native tissue from which they were derived with respect to cellular composition and architecture. For this reason, organoids are becoming the new gold standard for undertaking intestinal epithelial cell research. Unfortunately, their otherwise advantageous three-dimensional geometry prevents easy access to the apical epithelium, which is a major limitation when studying interactions between dietary or microbial components and host tissues. To overcome this problem, we developed porcine colonoid-derived monolayers cultured on both permeable Transwell inserts and tissue culture treated polystyrene plates. We found that seeding density and culture format altered the expression of genes encoding markers of specific cell types (stem cells, colonocytes, goblets, and enteroendocrine cells), and barrier maturation (tight junctions). Additionally, we found that changes to the formulation of the culture medium altered the cellular composition of colonoids and of monolayers derived from them, resulting in cultures with an increasingly differentiated phenotype that was similar to that of their tissue of origin.

Migrasomes are newly discovered, migration-dependent organelles that mediate the release of cellular contents into the extracellular environment through a process known as migracytosis. Since their identification in 2014, growing evidence has highlighted their critical roles in intercellular communication, organ development, mitochondrial quality control, and disease pathogenesis. Migrasome biogenesis is a complex, multi-step process tightly regulated by lipid composition, tetraspanin-enriched microdomains, and molecular pathways involving sphingomyelin synthase 2, Rab35, and integrins.

Developing efficient, low-cost catalysts for oxygen reduction and evolution reactions (ORR and OER) is key to advancing metal-air batteries and regenerative fuel cells. In this study, nitrogen-doped binary metal (Mn and Ni) oxides (N-BMOs) and Pt-decorated N-BMOs were synthesised using three methods and tested as ORR and OER catalysts in alkaline media. Their physicochemical properties were characterised by XRD, N-sorption, TEM, and XPS, while their electrochemical performance was evaluated using voltammetry and impedance spectroscopy.

Developing solid electrolytes with high ionic conductivity, a high voltage window, low flammability, and excellent interface compatibilities with both the anode and cathode for lithium-metal batteries is still a great challenge but highly desirable. Herein, we achieve this target through an in situ copolymerization of vinyl ethylene carbonate (VEC) together with acrylonitrile (AN) under fitting ratios inside a porous polyacrylonitrile (PAN) fiber membrane doped with flame-retardant decabromodiphenyl ethane (DBDPE) molecules. The received fiber-reinforced polycarbonate-based composite electrolyte with an ultrathin thickness of 13 μm exhibits good internal interfacial compatibility because of the same AN structure and superior flame-retardant performance due to the doped DBDPE molecules.
