Article Abstract

Total synthesis of and diastereomers of prenylated davanoids like davanone, nordavanone, and davana acid ethyl ester was achieved in an enantioselective strategy. Various other davanoids could also be synthesized using standard procedures from the Weinreb amides derived from davana acids. Enantioselectivity in our synthesis was achieved employing a Crimmins' non-Evans syn aldol reaction that fixed the stereochemistry of the C3-hydroxyl group, while the C2-methyl group was epimerized in a late stage of the synthesis. A Lewis acid-mediated cycloetherification reaction was used to establish the tetrahydrofuran core of these molecules. Interestingly, a slight alteration of the Crimmins' non-Evans syn aldol protocol led to the complete conversion of the aldol adduct to the core tetrahydrofuran ring of davanoids, thus essentially dovetailing two important steps in the synthesis. The resulting one-pot tandem aldol-cycloetherification strategy enabled the enantioselective synthesis of davana acid ethyl esters and 2--davanone/nordavanone in just three steps in excellent overall yields. The modularity of the approach will enable the synthesis of various other isomers in stereochemically pure forms for further biological profiling of this important class of molecules.

A one-pot strategy was developed for the synthesis of α-substituted 2-benzofuranmethamines from salicylaldehydes, phenylacetylenes, and cyclic secondary amines using CuFeO as a bifunctional catalyst. The reaction proceeds at 80 °C in 1,4-dioxane using CsCO as a base, enabling sequential A-coupling, 5--dig cyclization, and 1,3-allylic rearrangement in a single operation. Unlike previous methods, this protocol employs non-precious metal catalysts and mild reagents, operates under moderate conditions, and provides direct access to α-substituted 2-benzofuranmethamines in good yields (80-96%) with broad substrate compatibility.

Herein, we report an efficient and general one-pot multicomponent strategy, innovatively developed for the construction of δ,γ-unsaturated β-hydroxy nitrile scaffolds. Using readily available aldehydes and ketones as starting materials and TMSCFBr as a difluorocarbene precursor, the transformation proceeds via tandem Aldol condensation, a difluorocarbene electrophilic reaction, and hydrolysis. Mechanistic studies revealed that the difluorocarbene activates the oxygen atom of the α,β-unsaturated carbonyl group through electrophilic orientation, significantly enhancing the electrophilicity of the carbonyl carbon, thereby driving the reaction to proceed with high selectivity via a 1,2-addition pathway.

A Brønsted acidic ionic liquid (BAIL)-catalyzed one-pot tandem reduction of quinoline to tetrahydroquinoline (THQ) followed by reductive alkylation by the aldehyde has been demonstrated under mild reaction conditions with a shorter reaction time. This step-economical synthetic approach is suitable for late-stage functionalization of complex bioactive molecules. The reaction is highly chemoselective and tolerates a wide range of reducible-sensitive functional groups.

In the first part of this study, magnetic nanoparticles were decorated with folic acid (Vitamin B9) under ultrasonic agitation in water, resulting in heterogeneous nano-biocatalysts (FA@γ-FeO), which were characterized using different techniques, such as VSM, FT-IR, and FESEM. The efficiency of the as-prepared magnetic nanocatalyst was assessed in the synthesis of quinoxaline derivatives. We report on the synthesis, functionalization, and structural characterization of 11 novel dimethylarsinate-functionalized arsenomolybdates(V), [RAsMoO(OH){AsO(CH)}] (R = HO, CH, CH, CH, 3,5-(HOOC)CH, 4-FCH, 4-FCCH, 4-FCOCH, 4-BrCH, and 4-NCH) and [AsMoO(OH){AsO(CH)}], featuring a reduced hexanuclear {MoO} core, peripherally coordinated by three dimethylarsinate ligands and centrally functionalized with diverse organoarsonate, arsenate, or arsenite groups, including carboxylated, fluorinated, brominated, and azido derivatives. Synthesized via a simple one-pot aqueous method, the compounds were thoroughly characterized by single-crystal X-ray diffraction, thermogravimetric analysis, and elemental analysis in the solid state. Solution-phase stability was assessed by multinuclear (H, C, and F) nuclear magnetic resonance, while gas-phase behavior and fragmentation pathways were probed through electrospray ionization mass spectrometry, and tandem mass spectrometry (collision-induced dissociation).
