Article Abstract

Natural polymer hydrogels are widely used in various aspects of biomedical engineering, such as wound repair, owing to their abundance and biosafety. However, the low strength and the lack of function restricted their development and application scope. Herein, we fabricated novel multifunctional chitin/PEGDE-tannic acid (CPT) hydrogels through chemical- and physical-crosslinking strategies, using chitin as the base material, polyethylene glycol diglycidyl ether (PEGDE) and tannic acid (TA) as crosslinking agents, and 90 % ethanol as the regenerative bath. CPT hydrogels maintained a stable three-dimensional porous structure with suitable water contents and excellent biocompatibility. The mechanical properties of hydrogels were greatly improved (tensile stress up to 5.43 ± 1.14 MPa). Moreover, CPT hydrogels had good antibacterial, antioxidant, and hemostatic activities and could substantially promote wound healing in a rat model of full-thickness skin defect by regulating inflammatory responses and promoting collagen deposition and blood vessel formation. Therefore, this work provides a useful strategy to fabricate novel multifunctional CPT hydrogels with excellent mechanical, antibacterial, antioxidant, hemostatic, and biocompatible properties. CPT hydrogels could be promising candidates for wound healing.

A dual-drug-loaded γ-polyglutamic acid (γ-PGA) hydrogel was synthesized, integrating third-generation dendrimers (G3) as cross-linkers for the simultaneous encapsulation of hydrophilic doxorubicin (DOX) and hydrophobic camptothecin (CPT). This hydrogel, formed via an efficient EDC/NHS-mediated reaction, demonstrated a biocompatible, porous, and pH-responsive network. The hydrophobic core of G3 significantly enhanced CPT loading, while γ-PGA facilitated effective DOX incorporation.

The treatment of chronic wounds remains a significant challenge in the clinical field, and optimizing the treatment plan through visual monitoring of wound healing is an effective way to solve such problem. Herein, we propose a feasible strategy to construct a smearable C-P-T/mCQDs hydrogel for real-time monitoring of wound infection and healing status, through the synergistic combination of modified carbon quantum dots (mCQD), cellulose nanofiber, tannic acid, and polyvinyl alcohol. The hydrogel can be readily applied on the skin and rapidly forms a gel dressing through high-density hydrogen bonding, demonstrating exceptional mechanical robustness (tensile elongation: 600 %) and autonomous self-healing capabilities.

Protein therapeutics have emerged as an exceedingly promising treatment modality in recent times but are predominantly given as intravenous administration. Transitioning to subcutaneous (SC) administration of these therapies could significantly enhance patient convenience by enabling at-home administration, thereby potentially reducing the overall cost of treatment. Approaches that enable sustained delivery of subcutaneously administered biologics offer further advantages in terms of less frequent dosing and better patient compliance.

Oxidative stress is a major contributor to the difficulties in chronic wound healing. Although antioxidant hydrogels have been developed, they are still insufficient for addressing the entire chronic wound healing process. In this study, a lutein-loaded multifunctional hydrogel dressing (Lutein/CMC/PVP/TA, Lutein/CPT) with synergistic antioxidation properties was developed by hydrogen bonding and electrostatic crosslinking of tannic acid (TA) with carboxymethyl chitosan (CMC) and polyvinylpyrrolidone (PVP).

Skin cancer is the world's fifth most diagnosed malignancy and is increasingly occurring in young adults. The elevated morbidity and mortality of skin cancer are known to be highly correlated with its frequent recurrence after tumor excision. Although regimens such as chemotherapy and/or immunotherapy are often administered following surgical treatments, the patients may suffer from severe side effects, drug resistance, and/or high cost during treatments, indicating that the development of an effective and safe modality for skin cancer after surgery is still highly demanded nowadays.
