Neonatal sevoflurane anesthesia induces persistent cognitive deficits in mice through CypD-dependent mitochondrial impairment in parvalbumin interneurons.

Repeated neonatal sevoflurane anesthesia induces cognitive impairment in adulthood, but its neuropathological mechanisms remain unclear. Parvalbumin (PV) interneurons, which rely heavily on mitochondrial stability, are susceptible to anesthesia. Mitochondrial matrix protein Cyclophilin D (CypD) is involved in cognition by regulating the mitochondrial function.

The FK506 binding protein 51 (FKBP51) is a stress-modulating protein implicated in stress-related psychiatric disorders. While FKBP51 has been extensively studied in the brain and other tissues, its role in the adrenal gland, a key part of the stress response, remains unexplored. To investigate Fkbp5 gene expression and FKBP51 function in the adrenal gland, we examined expression in C57Bl/6 male and female mice and generated an adrenal cortex-specific Fkbp5 knockout (AS).

The human immunodeficiency virus 1 (HIV-1)-based lentivirus has been widely used for genetic modification. However, the efficiency of lentiviral-based gene modification in Madin-Darby bovine kidney (MDBK) cells is considerably limited. In this study, we have shown that siRNA-mediated depletion of , a restriction factor in HIV-1 infection, can dramatically enhance HIV-1 infection in MDBK cells.

The role of thyroid-stimulating hormone receptor (TSHR) in macrophages on adipose tissue metabolic disorders remains unclear. We generated macrophage-specific TSHR knockout mice (LysM-Cre Tshr) using the Cre/LoxP system and induced subclinical hypothyroidism (SCH) via methimazole treatment. Metabolic/inflammatory phenotypes were assessed via glucose/insulin tolerance tests, histology, and molecular analyses.

Unlabelled: Patients who are recipients of allogeneic transplants or have underlying autoimmune disease require immune suppression, often with calcineurin inhibitors (CNI). There is an expanding repertoire of immune effector cell (IEC) therapies, including CD19 CAR-T cells and viral-specific T cells (VSTs), deployed in these patients; however, ongoing CNI therapy may be detrimental to IEC function. We thus developed a CRISPR/Cas9-based approach to engineer dual CNI [cyclosporine (CsA) and voclosporin (VCS)] resistant IEC therapies by targeting (encoding cyclophilin A - CypA), a critical binding partner for both drugs.
