Extracorporeal photopheresis (ECP) is a well-established, safe, and effective immunomodulatory therapy currently used in clinics to decrease T cell-mediated immunity in various disorders, including autoimmune diseases and chronic rejection in organ transplantation. Although the ECP procedure has been shown to induce apoptotic cells that are reintroduced into the patient at the end of the treatment, the precise tolerogenic mechanisms mediated by ECP are not fully understood. Previous in vitro studies have demonstrated that early apoptotic cells express annexins on their cell surface, which suppress myeloid cell activation on stimulation with bacterial lipopolysaccharide through Toll-like receptors.

Background: Mesenchymal stem cells (MSCs) are one of the effective treatments for acute graft-versus-host disease (aGVHD) following allogeneic hematopoietic stem cell transplantation (allo-HSCT) due to their potent immunoregulatory function. Given the limited quantity and high heterogeneity of freshly isolated MSCs, extensive expansion is essential for clinical application. Prolonged passaging of MSCs leads to a decline in therapeutic efficacy, with the underlying mechanisms remaining unclear.

Multivalent interactions between lectins and glycans are crucial for biological recognition; however, predicting functional inhibition based on binding affinity remains challenging. Herein, we investigated a series of structurally defined N-acetylglucosamine (GlcNAc)-functionalized dendrimers (1a-1c and 2a-2c) to examine how spatial orientation and temperature influenced the inhibition of wheat germ agglutinin (WGA). Using enzyme-linked lectin assays (ELLAs), we observed biphasic inhibition profiles for all the dendrimers, characterized by an initial enhancement of WGA binding at low concentrations, followed by effective inhibition at higher concentrations.
