Article Abstract

Interleukin (IL)-1 receptor type 1 (IL-1R1) activation triggers a proinflammatory signaling cascade that can exacerbate kidney injury. However, the functions of podocyte IL-1R1 in glomerular disease remain unclear. To study the role of IL-1R1 signaling in podocytes, we selectively ablated podocyte IL-1R1 in mice (PKO mice). We then subjected PKO mice and wild-type controls to two glomerular injury models: nephrotoxic serum (NTS)- and adriamycin-induced nephropathy. Surprisingly, we found that IL-1R1 activation in podocytes limited albuminuria and podocyte injury during NTS- and adriamycin-induced nephropathy. Moreover, deletion of IL-1R1 in podocytes drove podocyte apoptosis and glomerular injury through diminishing Akt activation. Activation of Akt signaling abrogated the differences in albuminuria and podocyte injury between wild-type and PKO mice during NTS. Thus, IL-1R1 signaling in podocytes limits susceptibility to glomerular injury via an Akt-dependent signaling pathway. These data identify an unexpected protective role for IL-1R1 signaling in podocytes in the pathogenesis of glomerular disease. The present study establishes that activation of the receptor for interleukin-1 limits susceptibility to damage to the kidney glomerulus in preclinical mouse models by stimulating Akt signaling cascades inside the podocyte.

Alstoniaschines A‒I (1‒9), nine previously alkaloids sharing five different skeletons were obtained from the leaves of Alstonia scholaris. The structures and absolute configurations were established by their extensive spectroscopic data analyses, including NMR, HRESIMS, X-ray crystallography data, and theoretical ECD calculations. Compounds 1, 2, 3, and 9 exerted significant protective effect against oxidative stress and inflammatory damage of podocytes induced by high glucose, manifesting as the increase of superoxide dismutase, catalase, glutathione peroxidase, alongside the reductions of malondialdehyde, nitric oxide, lactate dehydrogenase.

Objectives: In this study, we explored the mechanism by which DDIT4 influences the polarization phenotypic transformation of macrophages and inflammation through the regulation of mTOR signaling pathway, providing a new mechanism and target for the treatment of diabetic nephropathy.

Methods: The degree of inflammation and injury in renal tissues of diabetic kidney disease (DKD) animal model was evaluated using biochemical assays, renal pathology examinations, and Western blot tests. Podocytes and macrophages were isolated from renal tissues to observe the extent of podocyte injury and the quantity and polarization phenotype of macrophage infiltration.

Background: Pulmonary arterial hypertension (PAH) associated with systemic lupus erythematosus (SLE) represents a significant complication with incompletely understood pathogenesis. Notably, podocytic infolding glomerulopathy (PIG), a rare pathological phenomenon in SLE-related renal lesions, has not been previously linked to PAH development. The cardiovascular safety profile of belimumab, a biologic agent for SLE treatment, requires continued surveillance, particularly regarding potential pulmonary vascular complications.

Ezrin, radixin and moesin regulate assembly of actin-based structures, link membrane-spanning proteins to cortical actin and are part of cell signalling hubs. The CLIC5A protein is very abundant in radixin-dependent inner ear hair cell stereocilia and in ezrin-dependent kidney glomerular podocyte foot processes and is essential for the structural integrity of these actin-based cellular projections. The functional relationship between ERM proteins and CLIC5A is incompletely understood and whether CLIC5A functions as a chloride channel is controversial.

Background: Podocyte loss and parietal epithelial cell activation are features of subtypes of glomerulonephritis and focal segmental glomerulosclerosis (FSGS). We recently reported that the podocyte-specific loss of Krüppel-like factor 4 (Klf4ΔPod ) triggers dysregulated glomerular STAT3 activation, podocyte loss with parietal epithelial cell activation and proliferation, leading to FSGS. While pharmacological systemic STAT3 inhibition attenuated this phenotype, it remains unclear whether the detrimental effects of Klf4 loss is primarily a result of dysregulated STAT3 activation intrinsically in podocytes.
