Purpose: As the mechanism of interaction between circular RNAs (circRNAs) and microRNAs (miRNAs) in regulating the development of prostate cancer (PCa) is not clear, this study focuses on investigating these effects.
Materials And Methods: Sample tissues were collected from the PCa of patients, and microarray analysis of human circRNAs was conducted. The expression of circ_0001686, hsa_miR-411-5p (miR-411-5p) were also detected by qRT-PCR. Circ_0001686 and miR-411-5p mimics were transfected into the PCa cell lines (CWR22RV1and LNCaP) and MTT, colony formation, Transwell, and scratch wound assays were used to analyze the biological behaviors of PCa cells. Si-circ_0001686 and ASO-miR-411-5p were used as negative controls, and dual-luciferase reporter assays were performed to verify the interactions among circ_0001686, miR-411-5p, and SMAD3/TGFBR2. The levels of SMAD3 and TGFBR2 in different treated PCa cells were measured by western blot, and experiments in a nude mouse model were carried out to strengthen the findings of miR-411-5p.
Results: The expression of circ_0001686 was up-regulated, while the expression of miR-411-5p was down-regulated in PCa cells. Moreover, circ_0001686 promoted cell proliferation, migration, and invasion. Molecular mechanism exploration revealed that circ_0001686 could reduce miR-411-5p, affecting the downstream target genes of SMAD3 and TGFBR2. and studies verified that miR-411-5p inhibits PCa progression.
Conclusions: Circ_0001686 can reduce miR-411-5p to increase the expression of SMAD3/TGFBR2, which consequently promotes the proliferation, invasion, and migration of PCa cells.

Aims: In-stent restenosis (ISR) is a significant limitation of coronary stent implantation, but the exact mechanism of ISR remains unclear. Patients after percutaneous coronary intervention (PCI) are in a hypercoagulable state; however, there is less information on its association with chronic coronary artery disease (CAD) in patients with ISR after PCI. We aimed to clarify whether or not CAD patients with ISR after PCI are in a hypercoagulable state and whether or not PS exposure on extracellular vesicles (EVs), blood cells (BCs), and endothelial cells (ECs) is involved in the hypercoagulable state.

Resistance-conferring mutations in the androgen receptor (AR) ligand-binding pocket (LBP) compromise the effectiveness of clinically approved orthosteric AR antagonists. Targeting the dimerization interface pocket (DIP) of AR presents a promising therapeutic approach. In this study, we report the design and optimization of -(thiazol-2-yl) furanamide derivatives as novel AR DIP antagonists, among which was the most promising candidate.

Background And Objective: Male fertility assessment through sperm morphology analysis remains a critical component of reproductive health evaluation, as abnormal sperm morphology is strongly correlated with reduced fertility rates and poor assisted reproductive technology outcomes. Traditional manual analysis performed by embryologists is time-intensive, subjective, and prone to significant inter-observer variability, with studies reporting up to 40% disagreement between expert evaluators. This research presents a novel deep learning framework combining Convolutional Block Attention Module (CBAM) with ResNet50 architecture and advanced deep feature engineering (DFE) techniques for automated, objective sperm morphology classification.

Prior research suggested the potential correlation between circulating immune cell phenotypes and prostate cancer (PCa). However, it remains unclear whether the correlation can be mediated by plasma metabolites. We performed a bidirectional 2-sample, 2-step Mendelian randomization (MR) study mainly utilizing the inverse variance weighted method to examine the causal role of circulating immunophenotypes on PCa and explore the mediation effect of plasma metabolites in the pathway from immunophenotypes to PCa.

Prostate cancer (PCa) is a type of malignancy that originates in the prostate gland, often characterized by uncontrolled cell growth and potential metastasis. Long non-coding RNAs (lncRNAs) play crucial regulatory roles in the progression of prostate cancer, potentially facilitating tumor growth and metastasis via mechanisms that involve the enhancement of aerobic glycolysis. This study aimed to investigate the functional role of lncRNA HANR in prostate cancer progression.
