Sesquiterpene synthases (STSs) catalyze the formation of a large class of plant volatiles called sesquiterpenes. While thousands of putative STS sequences from diverse plant species are available, only a small number of them have been functionally characterized. Sequence identity-based screening for desired enzymes, often used in biotechnological applications, is difficult to apply here as STS sequence similarity is strongly affected by species. This calls for more sophisticated computational methods for functionality prediction. We investigate the specificity of precursor cation formation in these elusive enzymes. By inspecting multi-product STSs, we demonstrate that STSs have a strong selectivity towards one precursor cation. We use a machine learning approach combining sequence and structure information to accurately predict precursor cation specificity for STSs across all plant species. We combine this with a co-evolutionary analysis on the wealth of uncharacterized putative STS sequences, to pinpoint residues and distant functional contacts influencing cation formation and reaction pathway selection. These structural factors can be used to predict and engineer enzymes with specific functions, as we demonstrate by predicting and characterizing two novel STSs from Citrus bergamia.

Azo dyes, prevalent in various industries, including textile dyeing, food, and cosmetics, pose significant environmental and health risks due to their chemical stability and toxicity. This study introduces the synthesis and application of a copper hydrogen-π-bonded benzoate framework (Cu-HBF) and its derived marigold flower-like copper oxide (MFL-CuO) in a synergetic adsorption-photocatalytic process for efficiently removing cationic azo dyes from water, specifically crystal violet (CV), methylene blue (MB), and rhodamine B (RhB). The Cu-HBF, previously available only in single crystal form, is prepared here as a crystalline powder for the first time, using a low-cost and facile procedure, allowing its application as an adsorbent and also serving as a precursor for synthesizing well-structured copper oxide (MFL-CuO).

Mechanochemical reduction of β-diketiminate (BDI) barium iodide precursors with K/KI resulted in the first barium inverse sandwich complexes containing the benzene dianion in yields of up to 54%. This most challenging isolation of highly reactive (BDI)Ba-(CH)-Ba(BDI) complexes, completes the family of heavier benzene inverse sandwich complexes and allows for a comparison of trends in the series from Mg, Ca, Sr to Ba. Syntheses, stabilities, structures, electronic states and reactivities of the full range are compared.

In this work, we developed nucleophilic 2-aminoallyl cations guided by DFT calculations. Computational insights enabled a comprehensive analysis of factors governing N-nucleophilicity and an accurate prediction of cycloaddition reactivity with electron-deficient alkenes. Notably, this study represents the first asymmetric [3 + 2] cycloaddition of amino-allyl cations with exocyclic double bonds, enabling the efficient synthesis of -pyrrolidines with excellent diastereoselectivities and enantioselectivities.
