Background: Cerebrovascular reactivity reflects changes in cerebral blood flow in response to an acute stimulus and is reflective of the brain's ability to match blood flow to demand. Functional MRI with a breath-hold task can be used to elicit this vasoactive response, but data validity hinges on subject compliance. Determining breath-hold compliance often requires external monitoring equipment.

Background: Non-obstructive azoospermia represents the most severe form of male infertility. The heterogeneous nature of focal spermatogenesis within the testes of non-obstructive azoospermia patients poses significant challenges for accurately predicting sperm retrieval rates.

Objectives: To develop a machine learning-based predictive model for estimating sperm retrieval rates in patients with non-obstructive azoospermia.

Half-life prediction of central nervous system (CNS) small molecules in humans using gradient tree boosting.

Future Med Chem

September 2025

Computational Science & Artificial Intelligence, Xenon Pharmaceuticals Inc, Burnaby, BC, Canada.

Aims: To develop a machine learning (ML) model for early-stage prediction of human half-life of oral central nervous system (CNS) drugs and to establish a curated dataset, including key and data, to support future modeling efforts.

Materials & Methods: Human and rat half-life, plasma protein binding (PPB), and liver microsomal clearance (LM) data for 76 diverse CNS drugs and candidates were obtained from public sources or evaluated at WuXi AppTec. Gradient tree boosting (GTB) models were constructed using ChemAxon's Trainer Engine.

Background: Automated cardiac MR segmentation enables accurate and reproducible ventricular function assessment in Tetralogy of Fallot (ToF), whereas manual segmentation remains time-consuming and variable.

Purpose: To evaluate the deep learning (DL)-based models for automatic left ventricle (LV), right ventricle (RV), and LV myocardium segmentation in ToF, compared with manual reference standard annotations.

Study Type: Retrospective.

The research team established a quantitative H NMR method to determine the relative ethoxy content (EO%) in ethylcellulose using a CDCl/TFA-d solvent mixture. High-field NMR spectroscopy enabled direct measurement without the use of internal or external calibrants by integrating the methyl proton signals (δ 1.15 ppm) and the methylene/methine proton signals (δ 2.
