Article Abstract

While plant polyphenols possess a variety of biological properties, exploration of chemical diversity around them is still problematic. Here, an example of application of the Ugi multicomponent reaction to the combinatorial assembly of artificial, yet "natural-like", polyphenols is presented. The synthesized compounds represent a second-generation library directed to the inhibition of β-amyloid protein aggregation. Chiral enantiopure compounds, and polyphenol-β-lactam hybrids have been prepared too. The biochemical assays have highlighted the importance of the key pharmacophores in these compounds. A lead for inhibition of aggregation of truncated protein AβpE3-42 was selected.

The catalytic asymmetric Mannich reaction is a multicomponent reaction which affords β-amino carbonyl compounds by utilizing an aldehyde, a primary or secondary amine/ammonia, and a ketone. β-amino carbonyl scaffolds are crucial intermediates for the synthesis of naturally occurring bioactive compounds and their derivatives. The synthesized natural compounds exhibit a broad spectrum of biological activities including anti-fungal, anti-cancer, anti-bacterial, anti-HIV, anti-oxidant, and anti-inflammatory activities.

Polysaccharides and polyphenols are major bioactive constituents of plant-based foods, and their efficacy is often modulated by intermolecular interactions. In this study, non-covalent binary complexes of Hovenia dulcis polysaccharides (HDPs) and quercetin were synthesized via molecular self-assembly. Structural characterization confirmed the successful non-covalent association of quercetin onto alcohol-precipitated HDP fractions-HDPs30, HDPs50, and HDPs70.

In this work, we report the design, synthesis, and application of a hyper-crosslinked heterogeneous organometallic porous organic polymer (Pd@TP-DPPF) catalyst for the efficient and sustainable dicarbofunctionalization of internal alkynes via a facile three-component reaction. This strategy enables the highly trans-selective syntheses of tetrasubstituted olefins in excellent yields. The catalyst is constructed by integrating triptycene (TP) and 1,1'-bis(diphenylphosphino)ferrocene (DPPF) into a robust palladium-based porous framework, resulting in a unique heterogeneous system that efficiently mediates the coupling of internal alkynes with readily available iodoarenes and aryl/methyl boronic acids.

Silver-Catalyzed Difluorocarbene Activation Enables Multicomponent Synthesis of δ,γ-Unsaturated β-Hydroxynitriles.

Herein, we report an efficient and general one-pot multicomponent strategy, innovatively developed for the construction of δ,γ-unsaturated β-hydroxy nitrile scaffolds. Using readily available aldehydes and ketones as starting materials and TMSCFBr as a difluorocarbene precursor, the transformation proceeds via tandem Aldol condensation, a difluorocarbene electrophilic reaction, and hydrolysis. Mechanistic studies revealed that the difluorocarbene activates the oxygen atom of the α,β-unsaturated carbonyl group through electrophilic orientation, significantly enhancing the electrophilicity of the carbonyl carbon, thereby driving the reaction to proceed with high selectivity via a 1,2-addition pathway.

Probiotics sourced from host-adapted microbes represent a sustainable innovation in aquaculture nutrition, aiming to replace antibiotics and enhance fish health. However, studies evaluating indigenous probiotic strains specifically adapted to the gut environment of target species remain limited. In this study, an indigenous strain was isolated from common carp () intestine, PCR-authenticated via dual endoglucanase amplicons (545 and 1311 bp), formulated as a powdered feed additive, and tested at 1, 10, and 100 mg/kg against commercial Nutri-Fish®, Infloran Bio®, vitamin C, and a supplemented control in an 8-week feeding trial with ( = 126) carp.
