Article Abstract

Heterozygous pathogenic variants in the gene result in several clinical features including impaired skeletal growth, ocular and otolaryngological abnormalities. Missense mutations in the triple helical region of the COL2A1 protein have been associated with lethal spondyloepiphyseal dysplasia (SED). In this study, we aimed to identify the underlying cause of a case of SED congenita (SEDC) in a 27-month-old child. A patient who was diagnosed initially with osteochondrodysplasia underwent a detailed clinical and radiological examination to obtain a conclusive diagnosis. The patient did not show any clinical features of hypochondrogenesis. Whole exome sequencing of the gene was carried out to identify the underlying genetic cause of the disorder. Variant annotation and filtration detected a heterozygous missense mutation c.1357G>A (p.G453S) in the exon 21 of the gene of the proband which was confirmed by Sanger sequencing. Neither parent carried the mvariant suggesting this was a new mutation. The mutation (c.1357G>A), identified in this case, results in more mild phenotype than other missense mutations in exon 21 which are known to cause lethal hypochondrogenesis. We showed, for the first time, that a missense mutation (p.G453S) in the triple helical region of the alpha 1 (II) chain of the COL2A1 protein underlies SEDC and is not always lethal.

Photophysical studies on the interaction of small molecules with various forms of nucleic acids are attracting attention nowadays in order to delineate the molecular level mechanism of various biological processes occurring in vivo. Herein, we employed vivid steady-state and time-resolved spectroscopic techniques to elucidate the detailed characterization of the binding interaction of a biologically active cationic dye thioflavin T (ThT) with double and triple helical forms of RNA - A.U duplex and U.

Helical nanographenes (NGs) play a crucial role in the development of chiral nanomaterials due to their distinctive optoelectronic and chiroptical properties. Herein, we report the efficient synthesis of two unprecedented azulene-embedded asymmetric triple helical NGs ( and ) with controllable helicene subunit lengths and π-extension. The crystallographic analysis confirms their highly twisted and asymmetric geometries.

Recombinant human collagen (rhCol) holds broad potential in biomedical and industrial applications due to its high purity and low immunogenicity. However, large-scale production of structurally stable and functionally active rhCol remains challenging. A novel strategy integrating collagen sequence optimization and microbial prolyl-4-hydroxylase (P4H) screening was developed to enable efficient production of triple-helical rhCol in Komagataella phaffii.

Prolyl-4-hydroxylase (P4H) catalyzes the hydroxylation of proline to hydroxyproline, a critical post-translational modification essential for the formation and stability of collagen's triple-helix structure. Although indispensable for recombinant human collagen (rhCol) production, P4H suffers from low expression levels, which significantly limit its industrial application. In this study, we achieved high-level expression of P4H in Komagataella phaffii (K.

Spins within molecules benefit from the atomistic control of synthetic chemistry for the realization of qubits. One advantage is that the quantum superpositions of the spin states encoding the qubit can be coherently manipulated using electromagnetic radiation. The main challenge is the fragility of these superpositions when qubits are to partake of solid-state devices.
