An undescribed substituted dihydroxanthene-1,9-dione, named funiculosone, was isolated together with its two analogues identified as mangrovamide J and ravenelin, from the culture filtrates of Talaromyces funiculosus (Thom) Samson, Yilmaz, Frisvad & Seifert (Trichocomaceae), an endolichenic fungus isolated from lichen thallus of Diorygma hieroglyphicum (Pers.) Staiger & Kalb (Graphidaceae), in India. Funiculosone was characterized, essentially by spectroscopic methods, as 4,8,9a-trihydroxy-3,4a-dimethyl-4a,9a-dihydro-4H-xanthene-1,9-dione. Its relative stereochemistry was deduced by single crystal X-ray analysis while the absolute configuration was assigned as 4S,4aS,9aS by ECD spectra in comparison to that of the closely related mangrovamide J. This latter, to which, not being an amide, an inappropriate common name was given, was only recently isolated, together with undescribed and known prenylatedindole alkaloids and chromone derivatives from an unidentified Penicillium sp. X-ray structural analysis of the isolated mangrovamide J, for which no biological activity was previously reported, revealed polymorphism and a new crystalline phase is described. All the compounds displayed antibacterial activity with an IC range 23-104 μg/mL when assayed against Escherichia coli Escherich and Staphylococcus aureus Ogston. Funiculosone also showed anticandidal activity against Candida albicans Berkhout with an IC 35 μg/mL.

Chlorinated hydrocarbons are widely used as solvents and synthetic intermediates, but their chemical persistence can cause hazardous environmental accumulation. Haloalkane dehalogenase from (DhlA) is a bacterial enzyme that naturally converts toxic chloroalkanes into less harmful alcohols. Using a multiscale approach based on the empirical valence bond method, we investigate the catalytic mechanism of 1,2-dichloroethane dehalogenation within DhlA and its mutants.

Background: The objective of this study was to compare the effects of daily consumption of white potatoes compared with white rice on cardiometabolic health in individuals with type-2 diabetes (T2D).
Objective: To determine the effects of white potato consumption compared to white rice (a commonly consumed refined grain) on indices of glycemic control and cardiovascular health in individuals with overweight or obesity and T2D.
Methods: In this randomized crossover study, comparative control trial, 24 adults with T2D [45-80 y, body mass index (kg/m) 25-40] consumed baked white potatoes (100 g) or calorie-matched white rice (75 g) daily for 12 wk, separated by a 2-wk washout, with assessments of glycemic control, lipids, inflammation, blood pressure, endothelial function, and body composition at baseline (only 1 baseline visit included as a covariate in statistical analyses), 6 wk, and 12 wk.

Mammalian ALOX15 are allosteric enzymes but the mechanism of allosteric regulation remains a matter of discussion. Octyl (-(5-(1-indol-2-yl)-2-methoxyphenyl)sulfamoyl)carbamate inhibits the linoleate oxygenase activity of ALOX15 at nanomolar concentrations, but oxygenation of arachidonic acid is hardly affected. The mechanism of substrate selective inhibition suggests inter-monomer communication within the allosteric ALOX15 dimer complex, in which the inhibitor binding to monomer A induces conformational alterations in the structure of the active site of monomer B.

Introduction: The Zika virus (ZIKV) envelope (E) protein is critical for viral replication and host interactions. Although glycosylation of the E protein is known to influence viral infectivity and immune evasion, the specific functional roles of E protein glycosylation in ZIKV infectivity in mosquito cells remain unclear.
Methods: In this study, we generated a deglycosylation mutant ZIKV with a T156I substitution in the E protein and investigated its effects on viral replication and viral-host interactions in mosquito C6/36 cells.
