Chan-Lam coupling is one of the most popular and easy methods to perform arylation of amines (N-arylations). This cross-coupling is generally performed by reacting aryl boronate derivatives with a variety of substrates involving nitrogen containing functional groups such as amines, amides, ureas, hydrazine, carbamates. This article summarizes the synthetic applications of this reaction and the efforts of scientists to develop novel and efficient methodologies for this reaction.

Sulfur(VI) fluoride exchange (SuFEx) compounds are gaining increasing attention due to their various applications. We present the Diels-Alder reaction of ethenesulfonyl fluoride and analogues to rapidly access cyclic and bicyclic SuFEx derivatives in moderate to good yields. These derivatives have been shown to be useful intermediates in a variety of synthetic transformations to expand the toolkit for the preparation of cyclic aliphatic sulfonyl fluorides.

Mitochondria-targeted photodynamic therapy (PDT) circumvents the short lifetime and action radius limitation of reactive oxygen species (ROS) and greatly improves the anticancer PDT efficacy. However, current approaches require different molecular engineering strategies to separately improve ROS production and introduce mitochondria targeting ability, which involve tedious synthetic procedures. Herein, we report a facile one-step cationization strategy that simultaneously improves the ROS generation efficiency and introduces mitochondria targeting ability for enhanced PDT.

Violacein exhibits antitumor activity, indicating potential for future clinical application. However, an efficient delivery system is required for the clinical use of this hydrophobic compound. Effective delivery systems can enhance the solubility and bioavailability of hydrophobic compounds like violacein, facilitating its clinical application for antitumor therapy.

Antigen-induced clustering of cell surface receptors, including T cell receptors and Fc receptors, represents a widespread mechanism in cell signalling activation. However, most naturally occurring antigens, such as tumour-associated antigens, stimulate limited receptor clustering and on-target responses owing to insufficient density. Here we repurpose proximity labelling, a method used to biotinylate and identify spatially proximal proteins, to amplify designed probes as synthetic antigen clusters on the cell surface.
