Despite the rapid discovery of genes for rare genetic disorders, we continue to encounter individuals presenting with syndromic manifestations. Here, we have studied four affected people in three families presenting with cholestasis, congenital diarrhea, impaired hearing, and bone fragility. Whole-exome sequencing of all affected individuals and their parents identified biallelic mutations in Unc-45 Myosin Chaperone A (UNC45A) as a likely driver for this disorder. Subsequent in vitro and in vivo functional studies of the candidate gene indicated a loss-of-function paradigm, wherein mutations attenuated or abolished protein activity with concomitant defects in gut development and function.

Background: Cerebellar pathologies in adults can have a wide range of hereditary, acquired and sporadic-degenerative causes. Due to the frequency in daily hospital, especially intensive care, settings, electrolyte imbalances are an important, yet rare differential diagnosis. The hypomagnesemia-induced cerebellar syndrome (HiCS) constitutes a relevant disease entity with clinical and morphological variability due to a potential progression of symptoms and a promising causal treatment.

In addition to the more famous canine parvovirus 2 (CPV-2), the Parvoviridae family includes other viruses able to infect dogs [canine chaphamaparvovirus (CaChPV), canine bocavirus-1 (CBoV-1), and canine bufavirus (CBuV)], whose etiological role is still controversial (mostly identified in animals with diarrhea but also detected in asymptomatic animals). The aim of this work was to evaluate the shedding of these common and recently discovered viruses in the dog population from the Campania region (Italy). A total of 170 feces from apparently healthy dogs were sampled and tested with specific real-time PCR.

Early weaning of piglets elicits weaning stress, which in turn induces oxidative stress and consequently impairs growth and development. Hydrogen-rich water (HRW), characterized by selective antioxidant properties, mitigates oxidative stress damage and serves as an ideal intervention. Objective: To explore novel therapeutic approaches for locally advanced cervical cancer (LACC), we evaluated the efficacy and safety of hyperthermia (HT) and/or targeted therapy combined with cisplatin-based concurrent chemoradiotherapy (CCRT).
Methods: This retrospective study analyzed 119 LACC patients (tumor diameter ≥4 cm) treated at our institution (Jan 2021-Oct 2024), stratified into: CCRT (n = 48), targeted therapy + CCRT (T-CCRT, n = 44), and HT + targeted therapy + CCRT (HT-T-CCRT, n = 27). Complete/objective response rates (CRR/ORR) at 3/12 months were assessed.
