Objectives: Ferritin is the major intracellular iron storage protein essential for maintaining the cellular redox status. In recent years ferritin heavy chain (FHC) has been shown to be involved also in the control of cancer cell growth. Analysis of public microarray databases in ovarian cancer revealed a correlation between low FHC expression levels and shorter survival. To better understand the role of FHC in cancer, we have silenced the FHC gene in SKOV3 cells.
Results: FHC-KO significantly enhanced cell viability and induced a more aggressive behaviour. FHC-silenced cells showed increased ability to form 3D spheroids and enhanced expression of NANOG, OCT4, ALDH and Vimentin. These features were accompanied by augmented expression of SCD1, a major lipid metabolism enzyme. FHC apparently orchestrates part of these changes by regulating a network of miRNAs.
Methods: FHC-silenced and control shScr SKOV3 cells were monitored for changes in proliferation, migration, ability to propagate as 3D spheroids and for the expression of stem cell and epithelial-to-mesenchymal-transition (EMT) markers. The expression of three miRNAs relevant to spheroid formation or EMT was assessed by q-PCR.
Conclusions: In this paper we uncover a new function of FHC in the control of cancer stem cells.

This report describes the case of a 43-year-old male presenting with severe anemia (hemoglobin, HGB 35 g/L). A comprehensive evaluation encompassing bone marrow morphology (24% ring sideroblasts), genetic testing (SF3B1-negative), iron metabolism studies (ferritin 1 179 μg/L), and imaging (liver MRI T2* value 1.1 ms) was performed for diagnosis, treatment planning, and assessment of outcomes.

Programmed cell death (PCD) refers to a regulated cellular process involving a cascade of biochemical reactions and molecular mechanisms, commonly including apoptosis, necroptosis, and pyroptosis. Ferroptosis is a recently identified form of PCD distinguished by its dependence on iron. Background: Idiopathic pulmonary fibrosis (IPF) is a chronic lung disease characterized by progressive scarring of lung tissue, which can lead to respiratory failure and other serious complications, making the search for safe and effective therapeutic agents an urgent priority; Maimendong Decoction (MMDD), a traditional Chinese herbal formula widely employed in clinical practice to treat pulmonary disorders, has been shown to reduce inflammatory cytokines, suppress pro-fibrotic factors, and alleviate oxidative stress, yet the precise mechanisms by which MMDD exerts its anti-fibrotic effects remain unclear.
Purpose: This study aims to evaluate the therapeutic efficacy of MMDD in pulmonary fibrosis and to elucidate its underlying mechanisms.
Methods: Using a bleomycin (BLM)-induced mouse model of idiopathic pulmonary fibrosis, we assessed the therapeutic impact of MMDD on pulmonary fibrosis; concurrently, Ophiopogonin-D (OP-D), a principal active ingredient of MMDD, was evaluated in an in vitro human fetal lung fibroblast model treated with transforming growth factor-β (TGF-β) to elucidate its precise anti-fibrotic mechanism; finally, multi-omics technologies and bioinformatics analyses were employed for comprehensive validation.
