Histone modifications reflect gene activity, but the relationship between cause and consequence of transcriptional control is heavily debated. Recent developments in rewriting local histone codes of endogenous genes elucidated instructiveness of certain marks in regulating gene expression. Maintenance of such repressive epigenome editing is controversial, while stable reactivation is still largely unexplored. Here we demonstrate sustained gene re-expression using two types of engineered DNA-binding domains fused to a H3K4 methyltransferase. Local induction of H3K4me3 is sufficient to allow re-expression of silenced target genes in various cell types. Maintenance of the re-expression is achieved, but strongly depends on the chromatin microenvironment (that is, DNA methylation status). We further identify H3K79me to be essential in allowing stable gene re-expression, confirming its role in epigenetic crosstalk for stable reactivation. Our approach uncovers potent epigenetic modifications to be directly written onto genomic loci to stably activate any given gene.

Purpose: To evaluate visual, anatomical and safety outcomes of aflibercept 8 mg in previously treated patients with neovascular age-related macular degeneration (nAMD).
Methods: This retrospective study included nAMD patients switched to aflibercept 8 mg from prior anti-VEGF therapies at Sahlgrenska University Hospital between February 2024 and February 2025. Data on best-corrected visual acuity (BCVA), central retinal thickness (CRT), pigment epithelial detachment (PED) height, fluid status, treatment intervals, time to fluid recurrence and adverse events were collected.

Microsatellite-stable (MSS) rectal adenocarcinoma remains a therapeutic challenge, particularly in patients with complicating factors such as chronic hepatitis B virus (HBV) infection. Advances in immunotherapy, including immune checkpoint inhibitors (ICIs), have introduced new opportunities to improve the treatment outcomes in this subset, yet their application in HBV-positive cancer patients is less well understood. Here we report the case of a 46-year-old female with MSS locally advanced rectal adenocarcinoma and active HBV infection, successfully treated with cmFOLFOXIRI combined with camrelizumab as neoadjuvant therapy.

Partial denitrification coupled with anammox (PD/A) has emerged as a promising low-carbon strategy for energy-efficient nitrogen removal from municipal wastewater. However, the reactivation of PD/A systems following operational disturbances remains challenging, particularly under continuous-flow conditions, where microbial interactions and process stability are more complex than in sequencing batch reactors. This study systematically and first evaluated the recovery dynamics of a continuous-flow PD/A process seeded with low-activity granular sludge stored at 4 °C for three months.

Background: Epstein-Barr virus-associated post-transplant lymphoproliferative disorder (EBV-PTLD) remains a life-threatening complication following allogeneic hematopoietic stem cell transplantation (allo-HSCT), especially in rituximab-refractory cases. Adoptive transfer of EBV-specific cytotoxic T lymphocytes (EBV-CTLs) offers a promising strategy to restore antiviral immunity, but long-term efficacy data, particularly in haploidentical transplant recipients, remain limited.
Methods: We conducted a retrospective study of 41 haploidentical HSCT recipients diagnosed with EBV-PTLD and received donor-derived EBV-CTLs.
