Neurokinin-2 receptor polymorphism predicts lymph node metastasis in colorectal cancer patients.

Article Abstract

To analyze the single nucleotide polymorphisms (SNPs) of two subtypes of neurokinin (NK) receptors, and (also known as and ), in colorectal cancer (CRC), peripheral blood samples were collected from 199 CRC patients. Direct-sequencing was performed to identify the rs10198644 and rs4644560 SNPs. Genotype results were correlated with clinical factors. The allele frequencies of rs10198644 GC, CC and GG were 52, 17 and 31%, respectively, while that of rs4644560 GC, CC, and GG were 36, 50 and 14%, respectively. Patients with rs4644560 GC exhibited more positive lymph nodes than those with CC (mean, 2.2 vs. 1.3; P=0.016). Further analysis highlighted that the number of positive lymph nodes was also increased in the rs4644560 GC/ rs10198644 GG group compared with the rs4644560 GG/ rs10198644 GG group (mean, 2.2 vs. 0.9; P=0.04). These data suggested that the rs4644560 GC polymorphism alone or combination with rs10198644 GG may be a promising prognostic marker of lymph node metastasis in CRC patients.

The regulation of follicular (F) and germinal center (GC) immune reactivity in human lymph nodes (LNs), particularly during the acute stages of viral infection, remains poorly understood: We have analyzed lung-draining lymph nodes (LD-LNs) from COVID-19 autopsies using multiplex imaging and spatial transcriptomics to examine the immune landscape with respect to follicular immune reactivity. We identified three groups of donors based on the Bcl6 prevalence of their Reactive Follicles (RFs): RF-Bcl6no/low, RF-Bcl6int, and RF-Bcl6high. A distinct B/TFH immune landscape, associated with increased prevalence of proliferating B-cell and TFH-cell subsets, was found in RF-Bcl6high LD-LNs.

Rosai-Dorfman disease mimicking IgG4-related disease.

Rosai-Dorfman disease (RDD) is a rare histiocytic disorder that may clinically and histologically resemble IgG4-related disease (IgG4-RD), especially in the presence of IgG4-positive plasma cell infiltration. In this case, a 69-year-old woman with generalized lymphadenopathy, constitutional symptoms, and elevated IgG4 levels was initially suspected to have IgG4-RD based on core needle biopsy. However, further evaluation with excisional lymph node biopsy revealed emperipolesis and S100-positive histiocytes, confirming the diagnosis of RDD.

Background: Mantle cell lymphoma (MCL) is a subtype of non-Hodgkin lymphoma characterized by the proliferation of B-cells. Severe hyperleukocytosis is an uncommon presentation that can complicate the diagnosis and management of MCL.
