A series of novel 3,3-dimethyl-2,3,4,9-tetrahydro-1H-carbazole derivatives were rationally designed, synthesized and evaluated for their biological activity as AcrB inhibitors. The compounds were assessed for their antibiotic potentiating effects, followed by evaluation of Nile Red efflux inhibition, and off-target effects including activity on the outer and inner bacterial membranes. Ten compounds potentiated antibiotic activity at sub-inhibitory concentrations, reducing the minimum inhibitory concentrations (MICs) of at least one of the tested antibiotics by at least 8-fold, with three derivatives (7c, 11g, and 11i) achieving 32-fold MIC reductions at 128 μg/mL.

A set of small molecules containing an unusual sp-rich heterobicyclic scaffold were prepared and evaluated in vitro for their ability to increase GLP-1 secretion in STC-1 cells and to protect SH-SY5Y cells from acute and chronic damage induced by glucose and methylglyoxal, respectively. The results obtained showed that some compounds, especially those containing an electron-withdrawing and/or lipophilic group at the meta position of the aryl moiety present at position 9, are effective non-covalent TRPA1 agonists. The lead compound so far individuated (compound 4b) was also prepared by asymmetric synthesis in both enantiomeric forms.

Perinephric myxoid pseudotumor of fat (PMPTF) is a recently characterized lesion typically associated with non-neoplastic renal disease. Its pathogenesis is thought to result from chronic renal "irritation," either due to mass effect from renal carcinoma or inflammation related to benign renal conditions. Objective: To describe treatment patterns for menopausal symptoms in women taking endocrine therapy for breast cancer treatment/prevention from the United States (US), United Kingdom (UK), and Germany.
Methods: We undertook a retrospective cohort study using data from the US Market Scan Commercial Claims and Encounters Data database, and electronic health records from the UK's Clinical Practice Research Database Aurum and the German Disease Analyzer. Women aged 18-65 years with a first prescription/dispensation for endocrine therapy for breast cancer treatment or prevention (index date) from 2010 to 2022 were followed up, and the following treatment classes were evaluated: antidepressants, benzodiazepines, anticonvulsants, antihypertensives, and hormone therapy.
