Article Abstract

Inflammatory bowel diseases (IBD) are highly heterogeneous conditions, varying in clinical manifestations, disease localization, progression, and response to treatment. Failing to account for this heterogeneity can substantially diminish the power of clinical trials and reduce the likelihood of detecting a true effect. In this review, we explore the transformative potential of Bayesian statistics in IBD clinical research, highlighting its ability to provide deeper insights, refine trial design, and facilitate more informed medical decision-making. We explain how Bayesian methods are best incorporated into innovative IBD clinical trial designs, such as single-arm trials utilizing historical data, master protocols, and adaptive trials. In adaptive designs, Bayesian techniques enable dynamic adjustments to sample sizes based on interim data, helping to maintain adequate power while optimizing resource allocation. For network meta-analysis, Bayesian statistics enhance the estimation of treatment effects in complex or sparse data situations by integrating prior knowledge and effectively managing hierarchical models. These methods are also applied in pharmacokinetic decision-making to address inter-patient variability in IBD, offering more accurate predictions of drug concentrations and target attainment at the outset of treatment. A checklist is added for non-specialist readers on how to approach reading an article that employs Bayesian methods, as part of a Users' Guide to the Literature.

The study of time-dependent within-person dynamics has gained popularity in recent years through the use of multilevel (latent) time-series models. However, due to the complexity of the models, model applications are usually limited with respect to the inclusion of time-varying moderating factors on the longitudinal within-person relations between variables. That is, in common applications of multilevel time-series models, the within-person dynamics of constructs over time are regarded as being insensitive to changes in other time-varying factors or changes in contexts.

Identifying causal genetic variants underlying economically important traits in dairy cattle is essential for understanding their genetic basis and optimizing breeding programs. The growing availability of sequenced reference genomes and individuals with both phenotypic and genotypic data notably enhances our ability to detect genetic associations and further pinpoint causal effects. Studying the association between mixtures of environmental exposures and health outcomes can be challenging due to issues such as correlation among the exposures and non-linearities or interactions in the exposure-response function. For this reason, one common strategy is to fit flexible nonparametric models to capture the true exposure-response surface. However, once such a model is fit, further decisions are required when it comes to summarizing the marginal and joint effects of the mixture on the outcome.

Background: Explore the changes in the burden of nasopharyngeal carcinoma (NPC) caused by occupational formaldehyde exposure from 1990 to 2021, and predict its development trend up to 2050, to provide references for formulating relevant policies and measures.

Methods: Using the Global Burden of Disease database 2021 (GBD 2021), we counted and analyzed the numbers and standardized rates of NPC deaths and disability-adjusted life years (DALYs) caused by occupational formaldehyde exposure globally, in 21 regions, and in 204 countries. Combined with correlation analysis, health inequality analysis, and frontier analysis, we further comprehensively described the disease burden and its changing trends.
